Objective To investigate the radioimmunoimaging and biodistribution of the 188 Re labeled McAb fragment HAb18F(ab′) 2 in human hepatoma xenografts in nude mice. Methods HAb18F(ab′) 2 was directly labeled with 188 Re by using 2mercaptoethanol as reducing agents. Labeling efficiency and immunoreactivity of 188 ReHAb18F(ab′) 2 were evaluated by Whatman 3mm paper chromatography and live cell assay respectively. SPECT with lowenergy generalpurpose collimator was selected for nude mice imaging after i.v. injection of 11.1 MBq of 188 ReHAb18F(ab′) 2. Biodistribution analysis was also conducted in nude mice bearing human hepatoma in which animals were killed and dissected after iv.v. injection of 1.85~259 MBq. Blood and tissues were removed, weighted and counted. Results The optimum labeling efficiency and immunoreactive fraction were 91.7 % and 078 respectively. Low energy generalpurpose collimator can resolve the problem of scattered counts. Imaging studies showed that the tumor of right lower extremities had retained the label at 3 h, and the optimal imaging result for tumor ranging from 20~30 h. After 45 h the radioation of whole body decreased. Biodistribution studies revealed that the label was concentrated specifically at tumor site, not concentrating at normal organs. The optimal imaging time was 20~30 h. Conclusion 188 Re labeledHAb18F(ab′) 2 can be uptaken selectively at the hepatoma site. This may provide a foundation for further study of the inhibition of tumor growth.$

目的 探讨混合裂变产物致癌的分子机理和混合裂变产物内污染诱发免疫细胞的放射免疫毒理效应。方法 应用碱性单细胞凝胶电泳技术观察了混合裂变产物急性染毒时对大鼠外周血淋巴细胞DNA的损伤。结果 裂片核素内照射对大鼠外周血淋巴细胞DNA具有明显的损伤作用 ,慧星细胞率和DNA的损伤程度随染毒时间的延长、累积剂量增大而逐渐加重 ,且存在较好的剂量 效应关系。结论 单细胞凝胶电泳技术检测的DNA单链断裂可作为毒物对健康影响的早期生物学指标。
