1981年5月京郊菠菜留种地发生菠菜矮花叶病。染病植株呈现畸形花叶,严重矮缩,影响抽薹结籽。通过病毒寄主范围的测定证明,它可侵染豆科、茄科、藜科、十字花科中的19种植物。可由桃蚜进行非持久性传毒。从提纯病毒的分析超离心得到三个沉降值,分别为57s、94s和113s。在电镜下观察到球形病毒颗粒,直径为24~25毫微米。根据现有资料分析,引起菠菜矮花叶病的病毒系为蚕豆萎蔫病毒的一个株系。

This paper reports results of the study on preparation of antiserum to Soybean Mosaic Virus and serological test with respective antiserum. SMV was purified from infected leaves, 15-30 days after showing symptoms by chloroform-butanol clearification, followed by polyethylene glycol (PEG) precipitation and ultracentrifugation. The ultraviolet absorption spectrum of the purified SMV was measured in a UNICAM SP 800 Spectrophotometer.

用差速离心法提纯6个大豆花叶病毒株,提纯液用紫外分光光度计扫描,以260mm的OD值计算病毒汁液浓度.影响提纯病毒浓度的因素有:(1)不同毒株同一提纯方法得率差异很大;(2)采病叶的时间:症状最明显时采叶提纯得率最高;(3)PEG处理时间以3.5小时较好. 将提纯病毒免疫兔子(以6号兔为例),共注射8.34mg,心脏采血,取得抗血清,通过微量凝聚、微量沉淀反应及琼脂扩散试验,在方法上进行了研究.
