The conditions of propagation of Mamestra brassicae nuclear polyhedrosis virus(MbNPV) in cell line Mb931104 were studied. The result showed that the conditions of TC-100 medium, a cell density of 3×10 5 cells/mL; and the cells of 3-day old were suitable for propagation of MbNPV. The conditional medium was not considered to be notably effect on the propagation of MbNPV.$

Methods The influences of culture liquid, passage, cell age and infective dose on the liter and yield of poliovirus were studied. Results The sensitivities of cells to poliovirus decreased with increasing age. The poliovirus yields per cell 24, 48, 72 and 96h after culture were 986, 745, 594 and 237 TCID50 respectively. The time span of CPE after infection with poliovirus was more than 48h. The viruses released to supernatant at the early stage of infection were unstable, and the infectious liters of them decreased by more than 0.25Log TCID50 every 24h. Conclusion The infectious liter and yield of poliovirus cultured in KMB17 cell are significantly influenced by culture conditions, such as nutrient componenl and cell age.

目的 为用KMB17细胞制备口服脊髓灰质炎减毒活疫苗提供适宜条件。方法 检测不同培养液、不同代次、不同细胞龄和不同病毒感染量等因素对病毒滴度及产量的影响。结果 细胞对脊髓灰质炎病毒的敏感性随细胞龄的增加而降低,培养后24、48、72和96h单个细胞中脊髓灰质炎病毒产量分别为986、745、594和237 Lo-.gTCID50;细胞感染脊髓灰质炎病毒后出现细胞病变时间跨度大于48h,早期从病变细胞释放出的病毒在培养上清中不稳定,感染性滴度每24h降低0.25 LogTCID50以上。结论 KMB17细胞培养脊髓灰质炎病毒的感染性滴度和产毒量受培养液营养成分和病毒接种时细胞龄等条件的影响。
