There are two main results. Firstly, we set up a (0,1)quadratic programming with linear contraints to simulate the construction of DNA restriction physical maps based on the experiements of cutting the DNA of larger viruses into many fragments. Secondly, we have designed cross network graph so as to construct and analyse the DNA restriction physical maps. Examples ars given to show the advantages of the approach.

From this library, 13 positive clones containing U2snRNA genes have been screened by in situ hybridization of bacteriophage λ plaques. The digestion pattern of recombinant DNA with several restriction endonucleases showed that there were 8 different clones found in 13 positive ones. One of the rice U2snRNA genes was located in a 1. 5 kb Sal I-BamH I fragment of the FDRGU2-3 clone.

水稻广陆矮4号黄化苗总DNA经Sau3A部分酶切,回收12~23kb片段,经CIP脱磷后克隆于EMBL3载体,建立了水稻基因文库.以拟南芥的U2.2snRNA基因为探针,从基因文库中筛选到13个阳性克隆,经不同酶切鉴定,初步判断有8种不同的克隆,对其中一个克隆FDRGU2-3的重组DNA构建了物理图谱,U2snRNA的一个基因已经定位在该克隆中1.5kb的Sail-BamHⅠ酶切片段上.

This paper reviewed the progress on the development of Becterial Artificial Chromosomes (BAC),construction approaches of physical mapping and strategies of performing targeted alterations such as site\|directed mutagenesis,deletions and replacement in BAC.Furthermore,the application of BAC transgenes in studying of the gene function,the phenotypic expression of an entire gene,or even distant regulatory sequences in their native sequence environment is also introduced.

本文综述了细菌人工染色体 (BacterialArtificialChromsomes ,BAC)的构建、物理图谱制作方法及其靶位精细操作策略的研究进展。同时 ,简要介绍了BAC转基因在基因功能以及基因表达与调控研究中的应用
