The paper is concerned with the isolation, purification and strucutral studies of hemicellulose in wheat straw collected from Gao You. Jiangsu. The wheat straw meal(40-60 mesh) was successively extracted with benzene/alcohol( 2:1 by volume), hot water and 0.5% ammonium oxalate to remove low molecular weight sugars, phenols, waxes and pectic substauces. The residue was delignified by treatment with sodium chlorite at room temprature for a duration of five days. Analytical data showed that a five-day treatment...

The paper is concerned with the isolation, purification and strucutral studies of hemicellulose in wheat straw collected from Gao You. Jiangsu. The wheat straw meal(40-60 mesh) was successively extracted with benzene/alcohol( 2:1 by volume), hot water and 0.5% ammonium oxalate to remove low molecular weight sugars, phenols, waxes and pectic substauces. The residue was delignified by treatment with sodium chlorite at room temprature for a duration of five days. Analytical data showed that a five-day treatment with chlorite yielded a holocellulose with a high yield of 88.1%, and only about 2.5% of the pentosan of the original straw was lost when the lignin content was reduced to 2.1%. It should be noticed that, when holocellulose was prepared by the Wise et al method, the losses of pentosan increased to 14% at the same lignin content. Evidently delignification of wheat straw at room temprature is more mild and more suitable for the subsequent structural study of hemicellulose than other methods. The resultant holccellulose was treated with 5% KOH. The total hemicellulose so obtained released throngh acid hydrolysis arabinose, xylose, mannose, galactose, glucose (in molar ratios of 1.22:10:0.05:0.14:0.21). The hemicellulose was then subjected to succesive precipitation with copper complex formed on the addition of Fehling's soluticn to an alkaline solution of the hemicellulose material. Through acid hydrolysis the acidic arabinoxylan released arabinose, xylose and traces of other sugars (in molar ratio of 0.96:10). The methoxyl content of xylan (1.09%) , corresponding to 4-0-methylglucuronic acids content 7.27%, Coped very well with uronic acid content determined with carbazole method (7.38%) ,suggesting the uronie acid in the xylan is 4-0-methylglucuronic acid. Molar ratios of arabinose, xylose and 4-0-nethylglucuronic acid is 1:10:0.6. The specific rotation of xylan is negative([α]_D-118.3°, in 2.5%NaOH aqueous), indicating that the xylan is of β-linkage. IR spectrum of xylan gave absorptions at 895 cm~(-1)., conforming very well results obtained by specific rotation. The xylan was converted to permethyl ether. The DP_n determined by vapour phase osmometry was ca.84. The methylated xylan was hydrolysed with formic acid and then with aqueous sulphuric acid. The hydrolysate, with some methylated sugars, were co?verted into alditol acetates. GC and GC-MS analyses inidicated the presence of al?itol acetates of 2,3,5-tri-0-methylarabinose, 2,3,4-tri-0-methylxylose, 2,3-di-0-methylxylose and 2-0-, and 3-0-methylxylose in molar ratios of 0.97:0.11:10:0.97. A sample of the methylated xylan was reduced with LiA1H, and then hydrolysed. The following sugars were identified by GC and GC-MS at the same conditions of their derived alditol acetates, 2,3,5-tri-0-methylarabinose, 2,3,4-tri-0-methylxylose, 2,3-di-0-methylxylose, 2(3-)0-methylxylose and 2,3,4-tri-0-methylglucose in the molar ratios of 0.92:0.09:10:1.37: 0.34. On oxidation with NaOI., 0.95 mole of periodate per sugar residue was consumed. The residual oxopolysaccharide, on acid hydrolysis, gave ethylene glycol, glycerol and traces of xylose. Neither methylation analysis nor periodate oxidation study gave any indication of the presence of main chain branches of xylan. It was concluded, there upon, that xylan possessed an average of ca.73 contiguous (1→4)linkage D-xylopyranosyl residues to which were attached ca.7 L-arabinofuranosyl residues and 4.4 4-0-methyl-D-glucopyranuronosyl residues at C-3 or C-2 positions of the xylan units.

A negative interrela-tionship existed between the content of hemicclkulose and winter food prefernces. Protein was sufficient for red deer, but energy might be a more eritical facter to wintering red deer.

The study indicates(1)the lignin content of the fibers is higher and hemicellulose content is lower than that of the parenchymas, (2)under identical conditions, the yield of the fiber pulp is higher and the alkali consumption is lower than that of the parenchymas pulp(3)the deligni-fication rate of the fibers is faster than that of the parenchymas. Both follow pseudo first-order kinetics. The removal of the hemicellulose in parenchymas is slower than that of the fibers.

本文首次建立了从麦草原料分离纤维细胞和薄壁细胞的简便有效方法,并比较了这两种细胎的Soda-AQ法制浆特性。研究结果表明:(1) 原料中纤维细胞的木素含量较薄壁细胞高,而半纤维素含量则是薄壁细胞较纤维细胞高;(2) 在相同的制浆条件下,纤维细胞的制浆得率较薄壁细胞高,而碱耗则是薄壁细胞的较高;(3) 纤维细胞脱木素速度较薄壁细胞的快,且都表现为假一级反应,薄壁细胞中半纤维素溶出较纤维细胞快。
