In the culture conditions in vitro, the seperated individual blastomeres developed eventually to tro-phectoderm vesicles without inner cell mass (ICM) though the developmental rate was high; the cell number was only 28.3±13.1, significantly lower than that of the blastocyst developed from fertilized ova. By nuclear transplantation, the formed embryos developed to blastocysts with ICM at evidently lower rate (28. 8%) than that of the fertilized embryos(75. 3% ,P<0. 01) ; but the cell number was similar (F>0. 05). It is suggested that the donor nucleus can be reprogrammed by the oocyte cytoplasm to a certain extent, if not completely. The low developmental capacities of nuclear transplant rabbit embryos are discussed.

本文就兔卵母细胞去核、胚胎细胞融合与卵母细胞激活等条件,对提高兔桑椹胚细胞核移植技术效率进行了进一步研究。结果表明,早龄卵母细胞(注射HCG后17~19h)更适合于做去核操作,平均去核率为75%,且所需移去的胞质少。用改进的新电融合液(含10mmol·L_(-1))组氨酸),操作卵的融合率(93.4%)、激活率(84.9%)得到明显提高。细胞松弛素B对融合胚激活及分裂并无有益作用。分离的供体胚单个卵裂球,尽管体外发育率很高,但最后均形成不含内细胞团的滋养层囊泡,所含细胞数(28.3± 13.1),显著地低于受精囊胚细胞数。而核移植胚胎囊胚(含内细胞团)发育率显著降低(28.8%),明显低于受精卵囊胚发育率(75.3%,P<0.01),但细胞数目相似(P>0.05)。结果提示,受体卵母细胞质似在某种程度上,对供体核基因表达(发育程度)进行了重新编排。文中还就核移植胚胎发育能力低有原因进行了讨论。

在原生质体培养再生植株成功的基础上,进行了体细胞杂交技术的研究,比较TPEG化学融合3种方法的融合效率及对细胞的毒害程度,发现在轻微摇动的情况下,于原生质体悬浮滴上逐渐加入PEG的方法效果最好。电融合试验结果表明:1.电融合以其高效、无毒害及简便易行等特点优于化学融合;2.应用国产的仅能输出尖波的电融合仪(DR—1型),利用渡金平行多电极(电极间距0,2mm,30个齿为一组交叉排列,总面积为18×18mm2),用1.1MHz,120—157峰-峰电压场强(Vpp/cm)的交变电场处理25~50秒,用1.25~1.50KV/cm的直流高压脉冲电击630μs左右,对大多数基因型可获得40%—80%的融合率和20%以上的细胞分裂率;3.在电融合液中加入高浓度的Ca2+对融合有明显抑制作用;4.融合效果与两亲本亲缘关系的远近有关。

The study was attempted to find the condition suitable for the electrofusion of protoplasts isolated from cell suspension cultures of Oryza sativa L. and Alternanthera philoxeroides (Mart.) Griseb. It showed that the fusion frequency of protoplasts reached 24.6% with the following parameters: alternate current (AC) 100V/cm , direct current (DC) 1.5kV/cm , DC pulse width (PW) 40μs , DC pulse interval 0.5 s, Number of DC pulse 2 , 10% mannitol and 0.4 mmol/L CaCl2 in electrofusion solution.

以水稻 (OryzasativaL . )的胚性悬浮细胞和空心莲子草 [Alternantheraphiloxeroides(Mart.)Griseb ]的悬浮细胞为材料 ,研究了两种原生质体电融合的条件。结果表明 :在交流电场为 10 0V/cm ,直流脉冲电场为 1.5kV/cm ,脉冲幅宽为 4 0 μs ,脉冲间隔为 0 .5s ,脉冲次数为 2次 ,电融合液组成为 10 %甘露醇和 0 .4mmol/LCaCl2 时 ,原生质体的融合频率达到 2 4 .6 %。
