An improved microanalytioal method for determination of serum high-density lipoprotein (HDL)-cholesterol is herein presented. 80 μl of serum in a small test tube was added to sodium phosphotungstate and Mg3+ mixed reagent. This solution was drawn up into a capillary tube, which was sealed and centrifuged at room temperature for 30 minutes for precipitation of both the serum low-density lipoprotein (LDL) and very low-density lipoprotein (VLDL). Cholesterol was then determined in the supernatant by means of improved O-phenyl-dimethyl-aldehyde method.This improved method is simple, rapid and replicable. Normal values obtained from 285 healthy adults determined by this method were found to correlate closely with those determined by other conventional methods(r = 0.84). In addition to the merits mentioned above, this method requires only drops of blood sample, no need of any special apparatus and could also be used for determining samples of hypertrigly-oeridemia. It is, therefore, recommended for clinical routine use and especially for large scale field surveys.
