    The cultural conditions and plant reproductive effect of apical meristem in vitro for diploid sugar beet were reported in this paper. Five media were used, the callus have got from all sugar beet apical meristem and reproduced plantlet on three media. The results indicated that the No.1 medium was optimum growth hormone for apical meristem culture,different types of growth substance and different concentrations of sugar have significant effect on inducing callus and differentiation rate of vitro-plantlet. The results indicated that the No.1 medium was optimum growth hormone for apical meristem culture,different types of growth substance and different concentrations of sugar have significant effect on inducing callus and differentiation rate of vitro-plantlet. The fit growth substance was NAA for the explant of apical meristem. The brawny root system can be induced with 1/2 MS composition plus NAA. The sandy soil were screened and sterilized with nutrient solutions which may increase establishment rate that transplanted from vitro to pot.

    本文报导了二倍体甜菜离休茎尖的培养条件和植株再生作用。采用5种培养基,由甜菜的茎尖组织都获得了愈伤组织,从其中的三种培养基上再生了植株。实验结果表明,不同种类的生长素,不同的蔗糖浓度对愈伤组织的诱导及绿苗分化率都有明显影响。对甜菜茎尖外植体,蒙乙酸(NAA)是适合的生长素。MS全成份的一半附加NAA可诱导出粗壮的根系。过筛消毒的砂土配合浇灌营养液可提高移栽苗的成活率。

    A filtrate from mycelial cultures of Verticillium dahliae was

    黄萎病菌培养滤液对陆地棉珂字201的愈伤组织及悬浮细胞系的生长和生活力有显著的影响。低浓度的病菌培养滤液,可抑制愈伤组织和悬浮细胞的生长,生活力下降;高浓度则可使细胞死亡。珂字201愈伤组织经诱变,在含10%~20%的黄萎病菌培养滤液的培养基上悬浮培养,经多次筛选获5个抗性细胞系。经鉴定,M2101015、M110101010和M12010三个细胞系在10%病菌培养滤液培养基上的存活率,由珂字201的34.85%提高到81.25%、85.00%和80.00%,且高于耐病品种爱SJ—1(63.52%)和SJ—5(56.67%);它们的原生质体对病菌培养滤液的敏感性降低。抗性细胞系植株再生研究表明,其胚胎发生能力均比对照下降,畸胚、畸苗增多。N11010的再生植株,经鉴定,对黄萎病菌培养滤液的反应是发病时间推迟,萎蔫程度明显减轻,表现一定的抗性。
