通过观察肾小管上皮细胞内Na+-K+-ATP酶活性、胞浆游离钙离子浓度及超微结构的变化,系统研究了重金属镉致肾小管细胞损伤的发病机理。结果表明,镉染毒后,肾小管上皮细胞内Na+-K+-ATP酶活性明显受抑,抑制率为82.4%,胞浆游离钙离子浓度较对照组明显增加(P<0.01),且伴有一系列超微结构损害。提示,镉致肾损伤时肾小管上皮细胞内Na+-K+-ATP酶活性受抑与胞浆游离钙离子浓度增加有明显的相关关系,胞内钙稳态失衡可能是镉致肾小管损伤的重要因素之一。

Objective:To investigate the cytosolic calcium concentration and its relevance to respiratory burst in lymphocytes from asthmatic children.Methods:The cytosolic calcium concentration 〔Ca2+〕i and superoxide generation in the peripheral lymphocytes were determined using Quin 2 as fluorescent indicator and the measurement of superoxide dismutaseinhibitable reduction of ferricytochrome C,respectively.Results:〔Ca2+〕i and O÷2 generation in lymphocytes from asthmatic children were significantly higher than those from healthy children.There were no significant differences in 〔Ca2+〕i and O÷2 generation in lymphocytes between intrinsic and extrinsic asthmatic children (P>005).〔Ca2+〕i in lymphocytes positively correlated with its O÷2 generation (r=041,P<005).Conclusions:The study indicates that easier activation of lymphocytes in asthma is partly due to the increment of 〔Ca2+〕i.The lymphocytes from asthmatic children produce more O÷2 when it is stimulated than those lymphocytes from healthy children,which may play an important role in the pathogenesis of asthma.

[Objective] To study the effects of niflumic acid (NFA) on the thrombocytic cytoplasmic free calcium [Ca2+]i and platelet aggregation rate (PAG) in diabetic patients. [Methods] Thrombocytic [Ca2+]i was detected by Fura-2 fluorescent technique, and PAG was detected by platelet aggregometer. We studied the effects of NFA, a chloride channel blocker, nifedipine, a calcium channel blocker, and their combining effects on thrombocytic [Ca2+]i and PAG in diabetic patients respectively. [Results] ① The PAG and [Ca2+]i in diabetic patients were significantly higher than that in control group. The platelet resting [Ca2+]i, Ca2+ release and Ca2+ influx in diabetic patients were higher than the control group. ② NFA and Nifedipine reduced the platelet Ca2+ influx which induced by thrombin and these effects were dose-dependent. The concentration producing 50% inhibition (IC50 ) of NFA was 50 μmol/L, the inhibition rate of thrombocytic [Ca2+]i was (54.7±14.5)%, and the inhibition rate of PAG was (32.3±21.4)% when the concentration of NFA was 50 μmol/L. The IC50 of Nifedipine was 7.5 μmol/L, the inhibition rate of thrombocytic [Ca2+]i was (17.9±11.9)% and the inhibition rate of PAG was (32.3±20.4)% when the concentration of nifedipine was 7.5 μmol/L. ③NFA(50 μmol/L) combining with nifedipine (7.5 μmol/L) inhibited platelet Ca2+ influx induced by thrombin in diabetes patients. The effects of NFA and nifedpine were independent. [Conclusions] Platelets in patients with diabetes are hyperactive. NFA reduces the PAG and platelet [Ca2+]i influx by blocking the chloride channels on platelet membrane in diabetic patients. There are no interactions between NFA and nifedipine on the movement of platelet [Ca2+]i.
