Objective To study the effect of α-crystallin on RGCs exposed to H_2O_2 and the possible mechanisms.Methods To observe the changes of RGCs exposed to 0.5mmol/L H_2O_2 in vitro during the period of intervention of α-crystallin at different concentrations(10~(-4)g/L,10~(-5)g/L,10~(-6)g/L) for 2h,RGCs was identificated by thy1.1 antibody.The number and the length of outgrowing RGCs axons were calculated under microscope and compared with the control group at 3,12,24h.The activity of SOD,CAT,GSH Px,LDH and the content...

Objective To study the effect of α-crystallin on RGCs exposed to H_2O_2 and the possible mechanisms.Methods To observe the changes of RGCs exposed to 0.5mmol/L H_2O_2 in vitro during the period of intervention of α-crystallin at different concentrations(10~(-4)g/L,10~(-5)g/L,10~(-6)g/L) for 2h,RGCs was identificated by thy1.1 antibody.The number and the length of outgrowing RGCs axons were calculated under microscope and compared with the control group at 3,12,24h.The activity of SOD,CAT,GSH Px,LDH and the content of MDA were detected by biochemistry technology.The apoptosis effect of RGCs was detected by DNA electrophoresis methods,The calcium ions level was detected by Laser scanning confocal fluorescence microimaging system.Results The survival rate and activity of SOD,CAT,GSH Px were decreased along with time in simple damage group,oppositely the activity of LDH and content of MDA were increased.α-crystallin group was opposite to simple damage group depending on concentration.The fragmentation of RGCs axons in 10~(-6)g/L α-crystallin group were less than them in simple damage group,there were apoptosis ladder-shaped band in the simple damage group at 24h point.There were weak apoptosis band in α-crystallin low concentration group.The calcium ions level was high in simple damage group,and 12h point achieved flat-top stage,but it decreased depending on concentration in α-crystallin group.Conclusion α-crystallin may reduce the free radical damage effect by H_2O_2 in RGCs.The mechanism might be involved in inhibiting calcium ions flowing into RGCs and increasing the activity antioxidant enzyme.

目的观察α-晶体蛋白(-αcrystallin)在过氧化氢(H2O2)损伤大鼠视网膜神经节细胞(retina ganglion cells,RGCs)中的影响并探讨其机制。方法原代培养大鼠RGCs并用Thy1.1鉴定,-αcrystallin组加入α-晶体蛋白(10-4、10-5、10-6g/L)预处理2h,然后加入H2O2(终浓度为0.5mmol/L)进行损伤实验,观察3、12、24h细胞形态学变化、轴突长度和突起数目的变化及细胞凋亡发生,检测RGCs培养上清液中相关酶(超氧化物歧化酶/SOD、谷胱甘肽过氧化物酶/GSH Px和过氧化氢酶/CAT、乳酸脱氢酶/LDH)活性及丙二醛/MDA水平的变化。Fluo-3/AM荧光比值成像技术测定RGCs胞浆游离钙离子水平。结果单纯损伤组RGCs存活率与SOD、CAT、GSH Px活性随损伤时间延长显著降低,LDH活性和MDA含量升高;-αcrystallin组RGCs存活率与SOD、CAT、GSH Px活性升高,LDH、MDA生成减少,其变化呈浓度依赖性。-αcrystallin组RGCs突起断裂程度明显少于单纯损伤组。单纯损伤组24h DNA电泳出现明...
