We have compared the conditions of producing phytase by Asp ficuum NRRL 3135 and Asp.niger 70 in solid state fermentation process.The results show that NRRL 3135 and ASN 70 all can use wheat bran as solid medium;and it would be beneficial for NRRL 3135 to have good pH control(with buffer),NRRL 3135 were incubated at 30±1℃ for 8~9 days,it can produce 5.2 u/g, but AsN 70 were incubated at 30±1℃ for5 ̄6 days,without buffer,it can produce 45.0u/g.

本文重点进行了无花果曲霉(Asp.ficuumNRRL3135)和黑曲霉(Aspniger70)用固体培养法生产植酸酶的对比研究。结果表明:NRRL3135与AsN70均可以麸皮为主要原料,NRRL3135用缓冲液(pH4.8)控制pH,在30℃±1℃培养8~9,产酶水平为5.2u/g干基;AsN70在不需要缓冲液控制pH的情况下,30℃±1℃培养5~6天,其产酶水平可达45.0u/g干基。

A phytase-producing strain separated from the mildewed solution of sodium phytate was studied. The basic medium of solid state fermentation (SSF) is wheat bran: corn meal: beau cake powder = 7: 2: 1. The optimal medium from optimization should be added 1% glucose and 4%(NH4) 2SO4 to the basic medium as well as the moisture is 150%. It shows from the dynamic data of SSFthat the degradation rate of phytate could be 90% at 28℃ after 6-d incubation, inorganic phosphorus and soluble protein are increased from 0....

从发霉植酸钠溶液中分离到一株产植酸酶的变灰青霉(Penicilliumcanescens)P4。以麸皮:玉米面:黄豆饼粉=7:2:1为主要培养基成份,用优选法确定最适培养基为在上述基本培养基中添加4%(NH4)2SO4,1%葡萄糖,1.5倍水,自然pH。发酵过程的动态分析表明,该菌在上述条件下28℃恒温培养6d后植酸降解率可达90%;无机磷含量由0.13%增至0.57%;可溶性蛋白含量由3.80%增至7.60%。用4%CaCl2·2H2O水溶液抽提固体发酵物,植酸酶活性可达312U/g干曲。

We got one strain Aspergillus niger(G2). G2 can produce 34u/g dry medium phytase in solidstate fermentation G2 suit for solid fermentation.

经大量菌种初筛和复筛,得到一株在固体发酵产植酸酶较高菌株G2,酶活达26千单位。G2菌株特别适合于固体发酵,并对植酸酶固体发酵酶活测定方法进行了探讨
