However, its application to secondary metabolites fermentation was hardly reported. In this study, fermentation broth was circulated through a dialysis device to eliminate detrimental metabolites from the broth, so as to enhance the productivity of spiramycin.Experimental results show that the specific production rate was maintain at higher level and the total yield of spiramycin was increased...

Membrane technology has been used chiefly to relief end product and toxicmetabolites inhibition during fermentation. However, its application to secondary metabolites fermentation was hardly reported. In this study, fermentation broth was circulated through a dialysis device to eliminate detrimental metabolites from the broth, so as to enhance the productivity of spiramycin.Experimental results show that the specific production rate was maintain at higher level and the total yield of spiramycin was increased by 30~50% with dialysis fermentation; besides, the fluctuation of growth and metabolic curves reduced evidently after applyng dialysis. Best yields were obtained under following conditions: membrane pore size,3.0μm; time for initiating dialysis, 60 h(after inoculation); ratio of working volume of culture uroth to dialysate, 1:1.

A reverse phase high-performance liquid chromatography was established for the analysis of meleumycin. Meleumycin was completely resolved to one another wither 10 minutes (such as culture liquid; adsorbent eluate; and intermediate etc) by means of a ODS-C_(18) column and a mobile phase of anhydrous methonal-0.2mol/L NH_4Ac-water, with UV detector at 232nm. Meleumycin A_1 was quantified. The calibration curve was linear in the range of 0.04-400μg/ml, average recovery was 102% and their coefficients of variation...

The feldback in potency of MLM among culture liquid and adsobent eluate was rapidly performed. A simple, easy, rapid and accurate method was provided for quality control of various kinds of intermediates. The result was quite satisfactory.

麦白霉素(Meleumycin,MLM)是放线菌发酵的次级代谢物,属于大环内酯类抗生素。在发酵工艺和菌种不同时都会产生一系列结构相似、性质相仿的同系物,其中的主要有效成分为A_1。本文建立了用反相HPLC法分析测定A_1的含量,及时反馈发酵液、吸附废液、解吸液、固体粗品中麦白霉素效价情况,为中间体各环节的质量控制提供了简便快速、重现性好的方法。使用Waters液相色谱仪,C_(18)柱(15 cm×7.5 mm),流动相为甲醇-NH_4Ac-水。

New Virginiamycin inducing factors were isolated from the culturebroth of Sfreptomyces antibioticus NF-18.They were named NFX-1,2,3 and 4,res-pectively.Among them,only NFX-2 and NFX-4,were obtained in enough quanti-ties to elucidate their structures on the basis of NMR,MS and IR spectra. Theyare 3, 8-dihexy1-4,9-dihydroxy-5,10-dimethy1-l,6-dioxadecane-2,7-dione(C22H40O6)and 3,8-(5′-methylhexyl)-4,9-dihydroxy-5,10-dimethy1-l,6-dioxadecane-2,7-diona(C21H44O6)for NFX-2 and NFX-4,respectively.
