In liquid medium,96% of the phytic acid was hydrolysed at 30℃ in 48 h by the organism. The production of phytase by the strain was affected by pH of the medium,fermentation time,fermentation temperature,composition of the medium and the aeration condition. The phytase was partially purified from the culture broth of Penicillium sp. P7 by using DEAE-52 chromatography and the characteristics of the phytase was studied. The enzyme showed an optimum pH of 5.0 and an optimum temperature of 60℃. The Km value of the enzyme was 0. 25 mmol/L.

从霉变的菜籽粕中筛选出具有产生植酸酶、降解植酸能力的青霉菌株P7。在液态发酵条件下,48h植酸降解率达96%。采用DEAE-52柱层析技术部分纯化植酸酶,对其生物学特征的研究表明:该植酸酶的最适反应温度为60℃,最适反应酸碱度为pH5.0,测得酶的米氏常数为0.25mmol/L。

Then the kinetic formula of cell growth rate, phytase production rate and glucose consumption rate were inferred by controling the specific biomass growth rate. The results of mathematic fitting between model calculated values and experimental values proved the validity and rationality of the kinetic models.

对黑曲霉 (AspergillusnigerND3 13 )液态发酵生产植酸酶的发酵过程和发酵动力学进行了研究。在摇瓶试验得到基本发酵工艺参数的基础上 ,应用 10L生物反应器进行放大试验取得了较好的结果 ,其最大菌体量 (干重 )达到 1 19g·10 0mL-1,酶活力达到 3 6u·mL-1。试验通过分批补料手段控制比生长速率的变化 ,得到了描述发酵过程的动力学模型并回归了模型参数。利用数学计算软件对试验数据与模型进行拟合 ,结果证明所建立的动力学模型能较好地反映并预测植酸酶的发酵过程
