Objective To explore the hemocytological and hemochemical changes in peafowls with infectious laryngotracheitis (ILT) after treatment with Chinese medicine S-anti-virus composite mixture. Methods Cytological and chemical examinations of the peripheral venous blood were conducted in ILT peafowls before and after treatment by daily oral therapy with Chinese medicine S-anti-virus composite mixture at the dose of 2 g/d. Results This Chinese medicine composite mixture increased the counts of RBC, HGB, and PCT, and decreased the activity of liver zymogram, and improved the renal functions in ILT peafowls. Moreover, serum IgM was up-regulated in ILT peafowls at 2 weeks and significantly at 4 weeks after treatment with this composite mixture. Conclusion The Chinese medicine S-anti-virus composite mixture complex has effects on hemostasis, anti-anaemia, recovery of liver functions, enhancement of immunological functions, and prevention of ILT in peafowls.

目的 探讨中药复方S 抗毒合剂治疗传染性喉气管炎 (ILT)后血液细胞和血液化学的变化。方法 以孔雀为实验对象 ,将中药按每日每只 2g拌饲料投喂ILT孔雀进行治疗 ,并于治疗前后取翅静脉血做血细胞和化学检测。结果 该药能提高ILT孔雀的RBC、HGB和PCT数 ,降低肝功酶谱活性 ,改善损伤的肾功指标 ,且在用中药 2周内使血清IgM升高 ;4周时IgA显著升高。结论 中药复方S 抗毒合剂对孔雀具有止血和抗贫血 ,恢复肝肾功能 ,提高机体免疫功能和防治ILT病的作用。

Aim To explore the differences of pathogenesis for the hepatotoxicity induced by chronic treatment of valproic acid(VPA) in different ages,and in combination administration with inducers of liver enzyme.Methods Animal models were established by oral administration chronically with VPA at doses of 200 or 500 mg·kg~(-1) per day in 30 days for 50 Wistar rats(infant and adult rats) with inducers of liver enzyme Phenobarbital(PB) or not.Mitochondria were obtained by differential centrifugation.Levels of liver enzymes,coagulation factors,plasma ammonia,VPA and PB serum levels,and L-carnitine in sera,as well as the changes of respiratory enzymes and lipid peroxidation in hepatic mitochondria were measured.Mitochondrial membrane potential(MMP) and mRNA expression of CYP450 reductase in liver were determined by flow cytometer and in situ hybridization,and morphological changes of hepatocytes were observed under microscope with Oil-Red-O staining.Results ① In all rats treated with higher dose of VPA added with PB or not,there were no significant elevations of liver enzymes(ALT and AST).However significant abnormalities of function of blood coagulation and serum fibrinogen were shown, and the levels of plasma ammonia and L-carnitine were also changed significantly,and the changes were notable in infant rats or in those rats added with PB. ② Average contents of cytochrome aa3 in liver mitochondria of infant rats were reduced by 58.80% and 61.80% because of administration of high dose VPA and high dose VPA added with PB,but were reduced by 37.55% and 46.53% in adults.As for activities of SDH,which affected by high dose VPA in infants,were significant decreased by 44.8% and 57.9%,respectively,but still in normal range in adult groups.Activities of CCO in liver mitochondria were significantly lowered by high dose VPA or added with PB compared with controls(P<0.01)in both age groups.Compared with controls,levels of glotathione-SH(GSH) in liver mitochondria were significantly decreased both in infant and adult rats treated with higher dose VPA or combinated with PB(P<0.01),whereas the levels of malondialdehyde(MDA) were significantly increased in both age groups(P<0.05).There was much severe in infants either GSH or MDA.③ MMP was reduced by 21.47% only in infant rats which treated with higher dose VPA added with PB and appeared significantly difference compared with controls(P<0.05).④ Mean optical densities(OD) on the expression of CYP450 reductase mRNA in infant rats with higher dose VPA were increased significantly compared with controls(P<0.05),and also enhanced remarkably in all rats coadministrated with PB.⑤ The morphological observation showed that significant lipid vacuoles were primarily accumulated in periportal areas of liver lobule in infant rats on frozen sections stained with Oil-Red-O,and the numbers of hepatocyte contained lipid droplets in infant rats was 7.5 times higher than that in adult rats in high dose VPA group,12 timeshigher in high dose VPA added with PB group,and there was significant difference statistically between the two age groups(P<0.01).Conclusions Presented study indicated that VPA can induce hepatotoxicity,and the dysfunction of liver mitochondria related with higher dose VPA,especially in infants and those combinated with inducers of liver enzyme.In addition,the increased expression of CYP450 reductase mRNA may be one of the significant mechanisms which main high risk factors can enhance VPA-associated hepatotoxicity.we should pay more attention to VPA-induced hepatotoxicity especially in infants and those which coadministration with inducers of liver enzyme,but liver enzymes(ALT and AST) are not a sensitive laboratory marker to VPA-associated hepatotoxicity.
