Objective: To evaluate the antipsychotic effects of olanzapine. Methods: The

目的:评价奥氮平的抗精神病作用。方法:通过苯丙胺诱导的小鼠定向运动实验、5-HTP诱导的小鼠头部抽动实验、大鼠僵住症试验,对奥氮平进行药效学观察,并与氯氮平及氟哌啶醇的药效进行比较。结果:奥氮平(ig,ED505.88mg/kg)、氯氮平(ig,ED5037.16mg/kg)、氟哌啶醇(ig,ED500.74mg/kg)能显著地拮抗苯丙胺诱导的小鼠定向运动,且有明显的量效关系。奥氮平(ig,ED500.54mg/kg)、氯氮平(ig,ED503.10mg/kg)、氟哌啶醇(ig,ED500.71mg/kg)能明显地拮抗5-HTP诱导的小鼠头部抽动。大鼠僵住症实验中,奥氮平、氯氮平、氟哌啶醇的ig,ED50分别是26,149,0.54mg/kg。结论:奥氮平有拮抗多巴胺及5-HT的活性,且其拮抗5-HT的作用比拮抗多巴胺的作用强,奥氮平的锥体外系不良反应轻,是一较好的抗精神病药物,奥氮平与氟哌啶醇合用对一些患者可能是较好的治疗方案。

Objective The purpose of this study was to observe the efficacy and safety of olanzapine in the treatment of schizophrenia Methods 65 patients with schizophrenia were included in this 6 weeks open trail clinical study The Positive and Negative Syndrome Scale (PANSS) and Brief Psychiatric Rating Scale (BPRS) derived from the PANSS were used to assess the clinical efficacy and the Abnormal Involuntary Movement Scale (AIMS), Simpson Angus Scale (SAS), and Treatment Emergent Side effect Scale (TESS) were used to assess the adverse effects Results Sixty two cases completed the 6 weeks treatment while 3 dropped out Among the 65 patients, 52% (34/65) met the criteria of complete relief, 28% (18/65) markedly improved, 8% (5/65) improved, and 12% (8/65) got no change The drug showed good effect on the positive, negative, as well as general psychopathological symptoms of schizophrenia Olanzapine showed low extrapyramidal symptoms during treatment The main adverse effects included anticholinergic effects, drowsiness, weight gain, and transient increase of liver enzymes, etc Conclusions The study suggests that olanzapine are a safe, effective antipsychotic agent with convenient dosing regime

目的 验证奥氮平治疗精神分裂症的疗效和安全性。方法 对65 例精神分裂症患者予以奥氮平在5~20 mg/d 治疗, 疗程为6 周。采用阳性与阴性症状量表以及简明精神病评定量表(BPRS) 评定疗效;采用不自主运动量表、SimpsonAngus 量表和副反应量表评定不良反应。结果 65例中脱落3 例( 在临床疗效评定中按无效计);完成治疗的62 例中完全缓解52% (34/65),显著进步28 %(18/65) ,进步8% (5/65) ,无效12% (8/65) 。奥氮平对阳性、阴性症状以及一般精神病态均有良好疗效。该药锥体外系不良反应少。主要不良反应为抗胆碱作用、困倦、体重增加、一过性肝酶活性升高等。结论 奥氮平是一种安全有效、服用方便的新型抗精神病药
