A new neutral proteinase-producing strain 0031 has been selected from the air of Hangzhou Leather Plant.In primary observation of the cultural property of the strain 0031,we had found that it was more resemble to Bac. subtilis(A.S.1398),but the enzymogram of fermentative liquor after disc electrophoresis was different from Bac.subtilis (A.S.1398). The adequate cultural conditions for the strain have been established: composition of the liquid medium(%)is:soya bean 3%,corn flocer 2%,wheat bran 3%,KH_2PO_4 0.03%,Na_2HPO_4...

The adequate cultural conditions for the strain have been established: composition of the liquid medium(%)is:soya bean 3%,corn flocer 2%,wheat bran 3%,KH_2PO_4 0.03%,Na_2HPO_4 0.4%,pH 8.0(before sterilization).After fermentation in shaking flasks(50ml/500ml/Δ,220 r.p. m.,ecc.dis.2.5cm.), at 28℃,for 38 hrs,the caseiaolytie activity was stablized at about 2000μg/ml, and the maximal activity could reach 2330μg/ml. During the process of study on the properties of enzyme-producing,we have found:1.suitable increase of aeration rate and pH,which will increase the enzymic produce and shorten the fermentative cycle;2.a tendency to slightly repress the enzymic produce and delay the enzymic peak,if fermentative temperature is above 30℃.3.no action of Zn■ and Ca■ on enzymic produce(test concen- tration ZnSO_4 0.005%,CaCl_2 0.3%).

A method described here is capable of direct visualization of protease zymogram on the disc electrophoresis gel. The method is more sensitive and convenient in comparison with the ordinary zymogram technique by which visualization is carried out on a substrate plate after contacting with the electrophoresis gel slab and enzymatic reaction. Polyacrylamide gel electrophoresis and enzymatic determination in vitro were used to demonstrate the relationship between hydrolase activity and resistance. The results are briefly stated as follows:(1) The earboxylesterase activity were considerably stronger in the resistant strain than in the susceptible strain. The decrease of Dipterex-resistance in the resistant strain reared in an insecticide-free environment was accompanied by a corresponding drop in earboxylesterase activity, and the earboxylesterase...

Polyacrylamide gel electrophoresis and enzymatic determination in vitro were used to demonstrate the relationship between hydrolase activity and resistance. The decrease of Dipterex-resistance in the resistant strain reared in an insecticide-free environment was accompanied by a corresponding drop in earboxylesterase activity, and the earboxylesterase activity in the resistant strain increased with the development of Dipterex-resistance.(2) Esterase zymograms from polyacrylamide gel electrophoresis revealed both strains had similar patterns except the E8 band which was much more intense in the resistant than in the susceptible strain, as judged by its hydrolysis of α-and β-naphthyl acetate. The results of eleetrophoresis scanning showed earboxylesterase increase mainly resided in this band. This band can also be found in the F1 strain (R × S) but the activity is less than in the resistant strain, which was in line with the decrease of resistance in the F1 strain.(3) No difference was observed in phosphatase and cholinesterase, not only in enzymatic determination in vitro but also in the esterase zymograms. The inhibition of cholinesterase of both strains by Dipterex and DDVP was similar.

本文利用离体水解酶测定和聚丙烯酰胺圆盘电泳技术研究了淡色库蚊(Culex pipiens pallensCop.)对敌百虫抗性和水解酶的关系。实验结果表明:(1)抗性品系的羧酸酯酶活力远比敏感的大,并随着对敌百虫抗性水平的升降而增减。(2)酯酶同功酶谱表明,抗性品系有一染色强度很深的特征性羧酸酯酶带E_5,该带在抗性和敏感品系的杂交子代F_1中也发现,但强度不如抗性品系,这同F_1代的抗性水平下降相平行。(3)无论胆碱酯酶或是磷酸酯酶活力,在二品系中未见到明显差异。抗性和敏感品系的胆碱酯酶对敌百虫和敌敌畏的敏感度相似。
