We have investigated the phenylalkylamine binding site in canine cardiac sarcolemmal preparations using (-)-[3H]-desmethoxyverapamil as the labeled ligand.
      
These data suggest the presence of a saturable, reversible and stereoselective phenylalkylamine binding site in canine cardiac sarcolemmal preparations which may be a receptor for the phenylalkylamine Ca++ channel inhibitors.

Electromicroscopically, all these materials showed the following dif-ferent structural changes in the myocardial fibres: 1) a conspicuous bul-ging of the sarcolemma and an active pinocytosis; 2) a cistern-like disten-tion of the interspaces between the double membranes of the intercalateddiscs; 3 ) multiplication and hypotrophy of the mitochondra and vacuolardegeneration; 4 ) disintergration, dissolution and disappearance of themicrofilaments; and 5)an increase in the wrinkles of the neuclear mem-branes. The Changes mentioned above indicate that the myocardium in thesecases is in a functional compensatory period and in some aged patientsslightly degenerative changes are found in the myocadium.

Weanling rats of both sexes were divided into two groups (10 rats in each group) ; the experimental group was fed on grains from Keshan disease endemic area,whereas the control group on grains from non-endemic area.By the end of three months of such feeding, all rats were sacrificed and the myocardial specimens for electron microscopic observation were prepared according to the methods described by Hoffstein et al.The abnormal La3+ deposition was observed between myofilaments and in some mitochondria in the rats fed on grains from Keshan disease endemic area.No obvious ultrasrructural injury could be found in the myocytes with abnormal La3 + deposition. In the control group,La3 + tracer was confined to the extracellular space.The result indicated that permeability of sarcolemmal membrane and some mitochondrial membrane in the rats fed on grains from Keshan disease endemic area was markedly increased, suggesting that alterations of myocardial cell membrane permeability may play an important role in the pathogenesis of the myocardial damage in Keshan disease.

When Mg2+ concentration is above 8 mM, it increases the activity due to Na++K++Mg2+. The Na++K++Mg2+/ Mg2+ activity ratio is therefore increased. Incubation of sarcolemma vesicles with high concentration Ca2+ leads to a decrease of the activating effect of either Na+ + K+ +Mg2+or Mg2+ alone. This inhibition of the enzyme activity...
