This paper reporias the formation and regeneration of Nocardia sp. protoplast. The research conditions are different from other aotinomyces because the cell wall of Nccardia isn't sensitive to lysozyme. The optimum conditions for efficient formation, purification and regeneration have been studied. First, Nooardia sp. was incubated with high concentration glyoine (3-4%). Then the mycelium was treated with mixed enzyme of lysozyme and proteinase B. P. or achromopeptidase, 108-109/ml protoplasts can be preparated....

The frequency of protoplast regeneration increased by addition of bovine serum albumn etc. When stored in refrigerator, protoplast was more stable in - 20℃ than in 4℃.

对诺卡菌sp.48原生质体形成和再生的各个方面作了较为详细的研究。由于诺卡菌在细胞壁组成上的特殊性,其原生质体研究的条件不同于其他放线菌。本实验用短小芽胞杆菌产生的胞外碱性蛋白酶B.P或消色肽酶与溶菌酶复合使用可分得10~8~10~9个/ml的原生质体,大大超过单用溶菌酶的效果。实验中摸索了甘氨酸的最佳浓度、制备原生质体的最佳菌龄及酶解作用的最适条件。结果提示,诺卡菌原生质体只能在自行设计的Rc再生培养基中再生,加牛血清白蛋白等有利于再生率的提高。原生质体贮存于-20℃比4℃更稳定。再生培养基中高浓度蔗糖影响菌落形态。还用相差显微镜和电镜分别观察了原生质体的形成过程及形态。

叔丁醇冻结干燥法用于植物材料,获得了良好的效果。经常规固定的样品完成脱水以后浸泡在叔丁醇中,然后将含醇样品放入电冰箱中冷冻,待叔丁醇凝固后再转移到真空镀膜机的钟罩里,用机械泵抽真空,冻结的醇在低真空中升华后样品得到了干燥。SEM 镜检表明,样品的表面和细胞断裂面及其深层(可见到内质网的双层膜、线粒体的嵴、高尔基体潴泡叠层、核糖体颗粒等细胞器)的三维图象均优于用临界点干燥法所得到的样品图象,叔丁醇冻结干燥法用于植物 SEM 样品制备,是一种操作简单,干燥质量可靠、值得推广的方法。

This paper describes comprehensively the moulding technological problems of AS boxes for fruits and vegetables used in refrigerator.The causes are emphatically analysed, and solving methods put forward.In the paper,the analytical theories and the applieable measures are available to all kinds of AS products.

本文就AS冰箱果菜盒成型工艺问题,着重分析其产生原因並提出解决措施。本文分析的理论和采取的措施对一切AS制品均适用。
