The change in activity of ornithine carbamyl transferase (OCT) in the liver of rats administered with carbon tetrachloride and 3'-methyl-4-dimethyl-aminoazobenzene (3'-Me-DAB) was investigated.The activity of OCT of the animals poisoned with carbon tetrachloride or fed with 3'-Me-DAB was found to be greatly reduced as compared with those of the control rats. In the former case, the activity was reduced 17% and in the latter, 35%. The combined effect of carbon tetrachloride and 3'-Me-DAB on the activity of the enzyme was found to be the most pronounced, with an activity scarcely 52% of that of the control. The OCT activity of the azo dye-fed rats showed a tendency to come back some time after administration of the drug was discontinued.The mechanism by which 3'-Me-DAB affected liver OCT activity is discussed.

The influence of iRNA, prepared from immunizing sheep with TA2 mouse mammary carcinoma cells, on the antitumor effects of hyperthermia and radiotherapy was studied in TA2 mice (high mammary carcinoma strain) with transplanted mammary carcinoma. The influence of iRNA, prepared from immunizing sheep with TA2 mouse mammary carcinoma cells, on the antitumor effects of hyperthermia and radiotherapy was studied in TA2 mice (high mammary carcinoma strain) with transplanted mammary carcinoma. Survival time of the mice following transplantation was 39.9 days for those treated with hyperthermia, 40 days with radiotherapy, 47.3 days with iRNA, and 23.5 days in the controls. The early tumor-growth inhibition rates (IR) for the three therapies were marked, the rates being 57.5%, 55.6% and 44.4% respectively. Hyperthermia in combination with iRNA yielded the survival time of 41 days and the IR 69.3%, whereas the combined effects of iRNA and radiotherapy were even better, the survival time being 63 days and IR 80.4%, It is suggested that radiotherapy combined with iRNA. preparation might be applicable in man.

本文用TA2纯系小鼠及其可移植性乳腺癌模型,研究了用该瘤免疫绵羊而提取的免疫核酸(iRNA)制剂对高温疗法、放疗抗肿瘤效果的影响,探讨联合应用治疗小鼠乳腺癌的可能性。实验结果表明:高温、放射及iRNA三者单独使用时,其平均存活期分别为39.3天、40天、47.3天,皆高于对照组(23.5天);前期抑制率分别为57.5%,55.6%和44.7%,示该三者均能明显抑制肿瘤生长。高溫与iRNA联合,生存期为41天,抑制率为69.3%,抗癌作用(与单纯高温相比)无明显增强。放射+iRNA生存期63天,抑制率80.4%,抗癌作用比其单独应用明显增强。本文对上述结果进行了讨论,并认为采用如放疗与iRNA联合治疗可能更为有益。

The effect of hyperthermia(43℃)and Pingyangmycin on human squamouscells carcinoma of tongue(Tca8113)has been investigated.All hyperthcrmic ex-periments(besides 37℃)were carried out in water baths at present tempcratures(±0.5℃).Domestic Pingyangmycin has been one of the most useful clinical anti-tumor drugs in oral maxillofacial surgery.The component of it is the same asBleomycin A5.When used as a low concentration Pingyangmycin(0.5μg/ml or5μg/ml for 1 hr.)was no significant cytotoxic effect for Tca8113 cells at 37℃.Although hyperthermia(43℃ for 1 hr.)alone had killing effect for part ofcells,the growth of most cells were inhibited temporarily.These cells may stillhave ability to growing and multiplication several days later.However the inre-versible synergic killing effect was observed when the cells were exposed toPingyangmycin at 43℃ for 1 hr.Furthmore,hyperthermia preceding Pingyang-mycin treatment produced a greater cytotoxic effect than Pingyangmycin treatmentproceding hyperthermia.We also demonstrated only little change in cytotoxicityabove 37℃ until a threshold of 42℃ to 43℃,when a synergistic interaction occ-Urs.Gradually enhancement rates of inhibiting ~3H-TdR up take at the temperaturerising,from 41℃ to 45℃,were shown in this report during combined hyperther-mia and Pingyangmycin.Using the phase contrast microscope and light microsc-ope,we observed the grade of cellular lesion not only at various temperaturewith Pingyangmycin but difference sequence by Toplin's morphologic classifica-tion.The results were something like as above experiments.The possible mecha-nism of combining effect of Pingyangmycin with hyperthermia was discussed.These results of the experiment may direct for clinical practice on thermo-chemotherapy for oral maxillofacial cancer.

本文选用细胞的生长曲线、集落形成实验、~3H-TdR 掺入试验并结合光镜下细胞受损程度的形态学观察等方法,用国产平阳霉素联合高温(43℃)对建株的人舌鳞状细胞癌 Tca8113系细胞进行体外实验,观察加温与平阳霉素是否具有协同杀伤作用。实验结果对口腔颌面部肿瘤的高温治疗,以及制定综合治疗方案有一定的指导意义。
