The locally selected drought-resistant cultivars, as compared with thosebred experimentally, have the advantages of higher respiration rate and highergermination energy and are able to maintain greater germination percentage andbetter root growth at a moisture stress as high as 18 atmospheres. It is suggested that in developing drought-resistant wheat varieties, such fac-tors as moisture intake capability, germination rate and ratio, root growth andrespiration intensity should be taken into consideration.

The germination of suspension culture pollens of Brassica napus L.cultivar 909 has been studied,by which we may use pollen as a tranmi-tter to introduce available gene.The main results are as follows:1.The most appropriate condition was that,in a fine day,atmospheric temperature is about 25℃,pollen were collected at 1∶00 p.m.andcultured at 20-25℃.2.The most proper concentration of saccharose for the germinationis 30%.3.Classic medium used for germination was adopted in suspensionculture(saccharose 30%,boric acid 120ppm).The highest germinationratio was only about 35%.4.If the medium was supplemented with 2.4-D,NAA,IAA,GA_3 or6-BA,KT,ZT 50ppm respectively,or with 2.4-D+GA_3,NAA+GA_3,IAA+GA_3,6-BA+GA_3,KT+GA_3,ZT+GA_3 50+30ppm respectively,thegermination would be promoted remarkably,especially the cytokinin plusGA_3,the germination ratio increased by more than 85%.5.Treat the pollens with the solution composed of 2.5% pectinase,1.5% cellulase(RS)and 30% saccharose,we isolated the pollen protoplas-ts successfully.

用909份甘蓝型油莱(Brassica napus L.)品种作试验材料,以花粉做媒介导入异种或同种有用基因,采用悬浮培养法,对花粉发芽进行了研究。结果:油菜花粉最适发芽的外界条件,是晴朗天气,气温25℃左右,午后1时取样,在温度25~30℃条件下发芽;花粉发芽最适的蔗糖浓度为30%;悬浮培养法,用通常的培养基(蔗糖30%,硼酸120ppm)进行发芽,最高萌发率仅在35%左右;普通培养基中附加2.4-D、NAA、IAA、GA_3各50ppm;或6-BA、KT、ZT 各50ppm;或2.4-D+GA_3、NAA+GA_3、IAA+GA_3、6-BA+GA_3、KT+GA_3、ZT+GA_3均为50ppm+30ppm 的不同生长激素处理,对花粉发芽均有显著促进作用。其中以细胞分裂素加 GA_3,促进发芽效果更佳,萌发率提高到85%以上;用2.5%果胶酶,1.5%RS 纤维素酶,以及30%蔗糖组成的酶液,成功地分离出油菜花粉原生质体。
