In the control group, the mean arterial pressure, cyclic GMP and atrial natriuretic factor were not influenced by the infusion of normal saline. These data suggest that the elevation of plasma cGMP is related to the hypotension and low circulatory resistance resulting from endotoxin shock. MB in vivo can effectively inhibit soluble guanylate cyclase and decrease plasma cGMP and increase mean arterial pressure of rabbits with endotoxin shock. MB may be an antagonistic to nitric oxide, suggesting that it can be used in treating septic shock.

Objective To characterize the blood lipid spectrum in subchronic arsenic poisoning rats and explore its possible mechanism.Methods Subchronic toxicity test was applied. Cyclic AMP、cyclic GMP and nitric oxide were detected in the liver homogenate and the light microscope and electron microscope preparations were made for certain liver tussue.Results ①In comparison with these in control group, the levels of TG and LDL in the serum of subchronic arsenic poisoning rats were decreased significantly (P<0.05) and a marked rise in the level of HDL was seen (P<0.05). The curve of dose effect of TC appeared a ‘r’ type.②The levers of cAMP and cGMP were significantly higher in the liver of arsenic poisoning rat than those in control group(P<0.05),and compared with that of control group ,the content of nitric oxide in the liver homogenate of arsenic poisoning rats was remarkably increased(P<0.05) and the dose effect relationship existed.③Under light microscope, in arsenic poisoning groups, infiltrating inflammatory cells were observed in liver portal area. Cells with lipid droplet inside and acidopilicity could also be seen in many foci. Ultrastructure change were as follows, there were plenty of lipid droplets inside the cells. Mitochondria and rough endoplasmic reticulum were swollen and the membrane damaged. Ribosome fallen off from rough endoplasmic reticulum.Conclusions Arsenic is able to influence the lipids metabolism. The balance of the second message was broken and the content of nitric oxide remarkably increased and the damage to the liver may play crucial roles in the abnormal lipid metabolism.

目的 观察亚慢性砷中毒大鼠血脂谱变化的特征并对其机理进行研究。方法 采用亚慢性毒性实验方法 ,大鼠经口灌胃染毒 ,实验期为 6周。实验结束后采血分离血清测甘油三酯 (TG)、总胆固醇 (TC)、低密度脂蛋白胆固醇 (LDL C)、高密度脂蛋白胆固醇 (HDL C) ,取部分肝脏左叶测一氧化氮 (NO)、环磷酸腺苷 (cAMP)、环磷酸鸟苷 (cGMP)并做光镜、电镜标本。结果 ①亚慢性砷中毒大鼠血清中TG、LDL C含量下降 ,与对照组相比有统计学意义 (P <0 .0 5 ) ,血清中HDL C含量与对照组相比显著升高 (P <0 .0 5 ) ,血清中TC的剂量效应曲线呈“γ”型。②染砷大鼠肝脏中cAMP、cGMP含量均显著高于对照组 (P <0 .0 5 ) ,且肝脏中NO含量明显升高 ,有剂量反应关系 ,与对照组相比差异有显著意义 (P <0 .0 5 )。③光镜观察可见染砷组肝汇管区内有炎性细胞浸润 ,肝细胞发生脂肪变性和嗜酸性变性 ;电镜观察发现肝细胞内有脂滴 ,线粒体肿胀 ,膜破坏 ,粗面内质网肿胀 ,脱颗粒。结论 砷可使脂代谢发生紊乱 ,出现这种情况可能与体内NO含量增多 ,第二信使系统平衡紊...
