In rats, atropine ( 4, 6, 10mg/kg ) , scopolamine ( 10mg/kg ) and B -7601 (15mg/kg) ip all caused a significant decrease in the concentration of plasma cyclic GMP (P<0.05), but the increase in the concentration of plasma cyclic AMP was insignificant (P>0.05) Atropine ( 6, 10mg/kg ) caused a significant increase of the ratio cAMP/cGMP (P<0.05) . The rank order of the potencies of these agents on plasma cyclic GMP was atropine>scopolamine>B-7601.

The effects of ginseng saponins on platelet aggregation, cyclic AMP and cyclic GMP level in platelets were studied. Ginseng saponins was found to be a potent inhibitor of platelet aggregation induced by arachidonic acid ( AA ) , ADP and thrombin in vitro and in vivo. The concentrations of ginseng saponins producing 50% inhibition of platelet aggregation induced by 0.55HM AA, 1.8-3.5uM ADP and 0.6-0.7 NIH u/ml of thrombin were 0.44, 1.20 and 1.32 mg/ml respectively. The effects of ginseng saponins on platelet aggregation, cyclic AMP and cyclic GMP level in platelets were studied. Ginseng saponins was found to be a potent inhibitor of platelet aggregation induced by arachidonic acid ( AA ) , ADP and thrombin in vitro and in vivo. The concentrations of ginseng saponins producing 50% inhibition of platelet aggregation induced by 0.55HM AA, 1.8-3.5uM ADP and 0.6-0.7 NIH u/ml of thrombin were 0.44, 1.20 and 1.32 mg/ml respectively. The inhibition % of aggregation induced by AA and ADP in vivo from 2-30min after iv administration of ginseng saponins ( 80 mg/kg) were 99-30.44 and 80.7-40.7, respectively. Also, cyclic AMP and cyclic GMP level in platelets were determined. It was observed that ginseng saponins significantly increased cyclic AMP level in platelets in a dose-dependent manner. But ginseng saponins did not affect cyclic GMP level in platelets. Our results indicate that the inhibition of ginseng saponins on platelet aggregation may be associated with an increase in cyclic AMP level.

本文报告了人参皂甙对家兔血小板聚集性,cAMP和cGMP含量的影响。结果表明,人参皂甙在体内外均抑制由花生四烯酸、ADP和凝血酶诱导的血小板聚集。同时,人参皂甙明显升高血小板中cAMP含量,但是不影响血小板中cGMP含量。提示人参皂甙对血小板聚集的抑制作用与其升高血小板中cAMP含量有关。

The values of cyclic AMP and cycic GMP in fasting plasma and 24-hour urine of 31 patients with urinary stone and of 20 normal control subjects were determined by radioimmunoassay so as to calculate the values of filtered, clearance and nephrogenous cyclic AMP and cyclic GMP.It was found that all parameters of cyclic AMP are significantly elevated in stone patients group. The values of cyclic AMP and cycic GMP in fasting plasma and 24-hour urine of 31 patients with urinary stone and of 20 normal control subjects were determined by radioimmunoassay so as to calculate the values of filtered, clearance and nephrogenous cyclic AMP and cyclic GMP.It was found that all parameters of cyclic AMP are significantly elevated in stone patients group. Stone patients have significantly lower-than-normal plasma cyclic GMP, significantly higher-than-normal clearance cyclic GMP and nephrogenous cyclic GMP. There are positive correlations betwesn plasma, urinary and nephrogenous cyclic AMP and cyclic GMP. It is supposed that the function of cyclic GMP is similar to that of cyclic AMP in the formation of stone.Some of the patients have primary or secondary hyper parathyroidism. Cyclic AMP and GMP are indirect parameters in the diagnosis of hyperparathyroidism in stone patients. Nephrogenous cyclic AMP or GMP and plasma cAMP/cGMP are more sensitive parameters. The values of urinary and nephrogenous cyclic AMP and cyclic GMP should be expressed as nmol/100mlGF.

本文采用放射免疫法测定了31例尿石症患者晨空腹血浆和24小时尿的cAMP和cGMP值,以此推算出滤过、清除和肾源性cAMP和cGMP值,并与20例健康人对照。结果发现尿石症者cAMP的几项指标均升高。血浆中cGMP降低,清除和肾源性cGMP增高,cAMP与cGMP值均呈正相关,推测cGMP在结石形成中的作用与cAMP相似。部分结石病人存在甲状旁腺机能亢进(简称甲旁亢),cAMP和cGMP可作为诊断尿石症患者甲旁亢的辅助指标,其中肾源性及血浆cAMP/cGMP值较敏感。测定值宜采用肾小球滤过率修正。
