CDKN_2 is a newly-reported tumor suppressor gene that controls cell cycle in direct pathway. But the report on the roles of CDKN_2 gene and its product, P16 protein, in the process of oral carcinogenesis is scarce, The results of this research showed immunohistochemically that the tissues of hyperkeratosis, premalignant lesions and invasive but nonmetastaticsquamous cell carcinomas had P16 expression. The positive staining appeared in cell nuclei and cytoplasms. And the positive degree and the heterogenecity of positive reactions trended to increase gradually with the development of cell maligancy. On the other hand,the positive degree in the tissues from patients with metastatic invasive squamous cell carcinomas was slightly lower than that of the other tissues and the positive stainingonly located in the cytoplasms of the primary lesions and their metastatic lymph nodes. It indicates that the P16 protein plays some roles in the carcinogenesis of human oral mucosa.

It had been unclear with regard to the roles of herpes simplex virus type Ⅰ(HSV-1)and the association between the infection of HSV-1 and tumour suppressorgenesuch asCDKN_2 or Rb in the process of oral carcinogenesis. This report demonstrated immunohistochemically that HSV-1 antigen expressed in each stage of oral carcinogenesis and the antigens existed in cytoplasms, The number and heterogenecity of positive cells increased gradually with the development of premalignant lesions. The expression of HSV-1 antigen related to the biological behaviors of invasive oral squamous cell carcinomas. It also revealed that the expression of1HSV-1antigen correlated with that of PRb but irrelated with that of P116. Through the method in situ hybridization it was disclosed that 44 percent of cases with HSV-1 antigens had positive hybridization signals of HSV-1 gene fragment, which appeared on the nuclear membranes. Theseresults suggest that HSV-1 may play an important role in the development of oral mucosal premalignant and malignantlesions by encoding some unknown proteins which can affect the functions of PRb and thus disturb the normal regulation of cell cycle.

Ⅰ型单纯疱疹病毒(HSV-1)在口腔粘膜癌前损害发生、发展过程中与抑癌基因的关系至今尚未明确。本研究应用免疫组织化学及组织切片原位杂交技术发现口腔癌变各阶段组织均表达HSV-1抗原,其阳性率无显著差异,HSV-1抗原主要在胞浆表达;随癌前损害的发展,在达HSV-1抗原的细胞数量增多,并出现阳性在达异质性;HSV-1抗原表达与鳞癌的生物学行为有关。相关分析表明,HSV-1抗原表达与P16无关,而与PRb相关。44%表达HSV-1抗原的组织细胞核膜存在HSV-1基因片段。这些结果提示,HSV-1可能通过其特定的蛋白产物与Rb等抑癌基因作用,影响细胞周期而参与口腔粘膜癌变的发生、发展。
