The optimum pH range and the optimum temperature of the immobilized cells were as same as that of the free cells. The batched hydrolysis of the immobilized cells were carried out every 24 hours for 30 days. The enzyme activity was stable and the average hydrolysis rate was 28.0%. The toxins were extracted by shaking after addition a mixture of methanol + water (3+1,V/V) as extractant and a mini-SAX solid phase column was employed for cleaning-up the sample. The toxins from cleaning up step were derivatized with O-phthaldialdehyde in the presence of mercaptoethanol as a catalyst to form highly fluorescent derivatives. A reversed...

The toxins were extracted by shaking after addition a mixture of methanol + water (3+1,V/V) as extractant and a mini-SAX solid phase column was employed for cleaning-up the sample. The toxins from cleaning up step were derivatized with O-phthaldialdehyde in the presence of mercaptoethanol as a catalyst to form highly fluorescent derivatives. A reversed phase LC was utilized for final separation of the sample and quantitation of the toxins was achieved by a fluorescent detection. The recoveries from fortified maize samples were found to be 78.0%— 95.7% with relative variation of 5.2%— 12.7 %, respectively. The minimum detection limits of the present method for FB1 and FB2 were 0.04 and 0.1 mg·kg-1, respectively.

本研究介绍了一种简单、快速分析测定玉米中腐马素毒素(fumonisins)残留的方法。样品用甲醇+水混合液提取,对提取液用小型强酸型阴离子固相萃取柱(SAX)纯化。纯化后的样品经化学衍生处理后,用反相高效液相色谱分离,并以荧光分光光度检测器定量测定。该方法对于添加已知量、添加水平在0.3—2mg·kg-1腐马素的玉米样的测定证实:其平均回收率在80%以上,而该方法对于腐马素三种同系物的最低检出限分别为0.1—0.05mg·kg-1。此外,本方法也对有限数目的大米、大麦、小米、高粱样中存在腐马素毒素污染的可能性进行了验证,结论是否定的。
