PURPOSE The changes on cell expansion and anti-tumor activity of mouse tumor-specific T cells activated by anti-CD3 monoclonal antibody(McAb) were investigated.The goal of this study is to find out more efficient method for tumor adoptive immunotherapy.METHODS Tumor-specific T cells from tumor-immunized mice were obtained by in vitro activation of anti-CD3 monoclonal antibody.The changes on cell expansion and anti-tumor activity as well as phenotype were studied.Four different culture modes were applied in this experiment:(1) cultured in anti-CD3 McAb alone(anti-CD3 group);(2) cultured in 20 000u/L rIL-2 alone(IL-2 group);(3) activated by anti-CD3 McAb for 48 hr and then cultured in 20 000u/L rIL-2 alone(CD3-IL-2 group);(4) activated by anti-CD3McAb for 48 hr and then cultured in CD3 McAb plus 20 000u/L rIL-2(CD3+IL-2 group).RESULTS The cells in CD3-IL-2 group and CD3+IL-2 group increased and expanded 780 folds and 610 folds respectively within 20 day.The maximum anti-tumor activities of both groups were 41.8% and 91.8% respectively on day 12.However,the continuous use of anti-CD3 McAb alone had neither proliferative effect nor anti-tumor activity.The IL-2group maintained a low level of proliferation within 20 days and showed its maximum anti-tumor activity of 30.2% on day 12.The study of cell phenotype by FACS analysis on day 6 indicated that>98% effective cells of both the CD3-IL-2 group and CD3+IL-2 group were Thy1.2+ cells.On day 20 Thy1.2+ cells of CD3+IL-2 group was still 98.8% while Thy1.2+ cell of CD3-IL-2 group was only 44.4%.There were also differences in CD4+ and CD8+ between the 2 groups.CONCLUSIONS The tumor-specific T cells cultured in the presence of the combination of anti-CD3 McAb and low dosed rIL-2 may facilitate a rapid expansion and long term maintenance of higher cytotoxicity.In general,this is a promising approach in tumor adoptive immunotherapy.

为寻找更有效的肿瘤过继性免疫治疗的方案,用α-CD3单克隆抗体(单抗)激活FBL-3红白血病肿瘤细胞免疫的小鼠脾细胞获取肿瘤特异性T细胞,并观察α-CD3单抗对肿瘤特异性T细胞的增殖、杀瘤活性及表型的影响。体外实验结果显示,免疫小鼠脾细胞经α-CD3单抗激活后,采用低剂量IL-2即可维持其长期高度的、持续的增殖;又培养过程中α-CD3单抗持续存在可大大促进其对肿瘤的特异性杀伤。细胞分型分析结果也支持了α-CD3单抗的持续存在对维持肿瘤特异性T细胞的抗瘤活性具有很重要的意义,是肿瘤过继免疫疗法中颇有前景的治疗方案。

Ntrogenor phosphorous-starved cells of the species were obtaned by growing the cells in f/ 2 media without addition of nitrogen or phosphorous unhl the ambient nitrogen or phosphorous had been depleted for 3 days. Such cells were used to study its uptake kinetics of NO3 -N NH4-N and PO4-P. In the NO3-N uptake experiments, the concentrations of 1, 2, 4, 6, 8, and 10 μmol / L were used. The same concentratons were adopted in the NH4-N uptake experiments. The concentrations used for the PO4-P uptake experiments were 0.5, 1, 2, 4, 6, and 8 μmol / L. The uptake rares were represented by the disappearing rates of the targeting nutrients. Semi-continuous culture mode was employed to examine the growth characteristics under the controlled nitrogen or phosphorous condihons. Five diluhon rates were employed, ranging between 0.05, 0.10, 0.20, 0.30, and 0.40. The concentfations of the limiting nitrogen and phosphorous in these experiments were 5 and 2 μmol / L, respechvely. The results show that in nutrient uptake experiments, the nutrient uptake nds (v) are constant withen the experiment Periods (0 to 60 min) and the nutrient uptake kinetics can be described by the Michaelis-Menton equation:v=Vm. s / (Ks+ s). The half saturation constants (Ks) for NO3-N NH4-N and PO4-P are 2.29, 2.18, and 1.40 pmol / L,respectively. Under the expenmenta condihons, maximal uptake rates (Vm) for NO3-N,NH-N and PO4-P are 7.28, 18.90, and 1.48×10- 13 mol / (cell. h), respectively. In the semixonhnuous cultllre experiments, the relahonship between steady state specific growth rates (μ) and cell quotas of the limited nutrient elements (q) follows the Droop equation:μ= μm.(1 - q / q0). Maximal growth rates Pm and minimum cell quota qo associated with the limited nitrogen concentrations are 1 .45d- 1 and 6.30×10- 13 mol / cell respechvely; those associated with the limited phosphorous concentrations were 1.37d- 1 and 0.69×10-13 mol/cell, respechvely. On the basis of the results obtained from the nutrient uptake experiments and the nutrient lindted growth exPeriments of P pungens, the speciflc maximal uptake rates for No3-N, NH4-N and PO4-P of this species are 28, 72, and 51d-1 respectively. Compared with the data of other phytoplanktonic species, P. pungens shows a high ability to upta-ke inorganic nitrogen, and a relatively low ability to uptake inonganic phosphorous. Therefore,phosphorous might be the controlling nutrient of this species in the natural environment.

于1991年11月在胶州湾采集尖刺拟菱形藻,以毛细管法分离纯化,建立克隆藻株,以此为实验材料,研究其在营养盐饥饿状态下对NO3-N,NH4/N和PO4-P的吸收动力学及在营养盐限制下的增殖特征。结果表明;在营养盐饥饿状态下,该藻对NO3-N,NH4N和PO4-P的吸收过程符合米氏方程,半饱和常数(KS)分别为2.29,2.18和1.40μmol/L。最大吸收速率(Vm)分别为728,1890,1.48×10-13’mol/(cell·h)。在营养盐限制下,该藻的比生长率与限制性营养元素细胞含量之间的关系符合Droop方程。求得该藻氮限制下的最大比生长率为1.45d-1,最小细胞氮含量为630×10-13mol/cell;磷限制下的最大比生长率为1.37d-1,最小细胞磷含量为0.69×10-13’mol/cell。根据以上结果,计算出该藻对NO3-N,NH4-N和PO4-P的比最大吸收速率分别为28,72和51d-1。研究说明,该藻具有较强的吸收氮的能力,而吸收磷的能力较弱,推测磷可能是自然环境中限制其生长的营养因子。

Based on the formation kinetics of monoclonal antibody,the hybridoma cell culture in bioreactor was optimized by the application of perfusion culture mode,the supplementation of potassium acetate,and the fortification of nutrients.When the bioreactor was operated with the daily perfusion of medium of 1/2 work volume,viable cell density in bioreactor was ,in comparison with batch culture,increased from 4.5×10 5cells/ml to 11×10 5cells/ml and antibody concentration in supernatant from 19mg/L to 28mg/L.After supplemented with 1g/L potassium acetate,the cell density kept constant and antibody production was improved to 38mg/L.The fortification of nutrients such as amino acids and vitamins raised the cell density to 42×10 5cells/ml and ,responsively,antibody concentration to 94mg/L.

根据杂交瘤细胞培养中单克隆抗体生产的动力学原理,采用灌注培养方式、添加醋酸钾和丰富营养物培养的途径,对反应器放大培养过程进行了优化。每天灌注1/2反应器工作体积,与分批培养相比,细胞密度由45×105/ml提高到11×105/ml,单抗浓度由19mg/L提高到28mg/L;添加1g/L醋酸钾,细胞密度基本保持不变,但单抗浓度增加到38μg/ml;用丰富营养物培养后,细胞密度和单抗浓度分别进一步提高到42×105/ml和94mg/L。抗B型红细胞单抗的血凝滴度,由分批培养的1:32,最终提高到1:256。
