The characteristics and system of a new kind of biosensor, fluoto-immuno sensor have been studied. In this paper reaction mechanisms of surface sensitive film are theoretically analyzed and the theoretical value of detective ensiivity and dynamic range of the sensor are given. The detective system is built and the sensing function is overall examined. Several properties of the sensor are discussed including binding and eluting of suiface antigen with antibody adsorption of non-specific protein, the influence of the adsorpton to the functions of the sensor and the reuse of sensor. Finally the testing results of the detective dynamic range is givea. The results correspond with the theoretical values.

本文较详细地分析了荧光免疫传感器表面膜的作用机理,给出了传感器检测灵敏度与动态范围理论值,建立了传感器检测系统,讨论了表面免疫反应的时间特性及非特异性蛋白吸附作用、吸附的消除及对传感器性能的影响,并比较了两种解离剂对表面膜使用重复性的影响。最后给出了传感器检测动态范围的实验数据,结果与理论值一致。

The determination of cholesterol is very important in clinical analysis. In this thesis, the development of a cholesterol oxidase biosensor is reported.Nylon mesh activated with dimethyl sulphate and reacted with lysine and glutaraldehyde, then cholesterol oxidase was reacted with GA and was immonilized on the nylon mesh. The determination of cholesterol is very important in clinical analysis. In this thesis, the development of a cholesterol oxidase biosensor is reported.Nylon mesh activated with dimethyl sulphate and reacted with lysine and glutaraldehyde, then cholesterol oxidase was reacted with GA and was immonilized on the nylon mesh. The nylon mesh with immobilized enzyme was fixed on an oxygen electrode. The optimized working conditions of the biosensor is : O.lmol/L phosphate buffer (pH6.3) as carrier solution, with a rate of 0.15-1.14mg/mL and the linear range is 0.25-1,00mg/mL, the slope is about 0.08μA.mL/mg. The response time of the sensor in linear range ≤2.5min and the life time is 15-day. This sensor has some advantages over those reported previously in working ranges and linear ranges.

本文初步研制了一种测定游离胆固醇的酶传感器。该传感器用于流动注射法的最佳工作条件:磷酸盐缓冲液作载液(pH=6.3,0.1mol/L),流动速度为0.5mL/min,温度为27℃,检测限为0.15mg/mL~1.14mg/mL,线性响应范围0.25~1.00mg/mL,斜率0.08μA·mL/mg,响应时间为0.5~2.0min,工作寿命15d。已将该传感器用于样品分析,取得初步满意的结果。
