Methods: After the model of transplanted pancreatic cancer was established innude mice with orthotransplantation of human pancreatic qqncer cell line into the pancreas, the ischemia ofthe right lobe of the pancreas was induced with ligation of the gastroduodenal, inferior pancreaticoduodenaland dorsal pancreatic arteries. The effects of the regional ischemia on the populatiDn proliferative and cytocycle kinetics on the pancreatic transplanted cancer were investigated. Results: The.transplanted cancer grewslower and its doubling time Was longer in the ischemic group than in the control. On the 3rd, 7th and l4thday after operation, the size of transplanted cancer, the proliferative index and protein content of the cancercells were significantly lower in the ischemic group than in the control (P<0. 01). ConcIusion: Reghnal ischemia can effectively inhibit the growth of transplanted pancreatic cancer in nude mice and suppress fhe sgnthesis of DNA and protein in the cancer cells.

目的:为缺血疗法在胰腺癌的临床应用提供基础研究资料。方法:在建立人胰腺癌细胞株裸小鼠胰腺原位移植瘤模型的基础上,结扎荷瘤棵鼠胃十二指肠动脉、胰十二指肠下动脉及背胰动脉诱导胰腺右叶区域性缺血,观察缺血对裸小鼠胰腺移植癌群体增殖动力学及细胞周期动力学的影响。结果:缺血组移植癌生长缓慢,倍增时间延长,移植瘤体积、癌细胞增殖指数及蛋白质含量术后3、7、14d均显著低于对照组(P<0.01)。结论:区域性缺血能有效地抑制裸小鼠胰腺移植癌的生长,并抑制癌细胞DNA及蛋白质的合成。

Objective To provide experimental data for the clinical application of ischemia therapy in treating pancreatic cancer.Methods The pancreatic transplanted cancer was obtained by transplanting human pancreatic cancer cell line PC-3 into right lobe of the pancreas in nude mice.Regional ischemia of right lobe of the pancreas was induced with ligating gastroduodenal,inferior pancreaticoduodenal and dorsal pancreatic arteries of the cancer-bearing animals.Effects of ischemia on proliferative kinetics and pathomorphology of the pancreatic transplanted cancer of nude mice were investigated.Results The transplanted cancer of ischemia group grew slower and their doubling time was longer as compared to the control.On the 3rd,7th and 14th days after operation,the volume of transplanted cancer,the proliferative index and protein content of the cancer cells in ischemia group were significently lower than those of control(P<0.01).Optical microscopy revealed large areas of coagulative necrosis,necrobiotic cells,inflammatory miflammatory cells proliferation and fibrosis infiltration were seen in ischemia group.The atrophy of acini,fibrosis and the infiltration of lyphocytes were found in peripancreatic tissue.Conclusions Ischemia can destroy and inhibit the pancreatic transplanted cancer of nude mice effectively.Establishment of the animal model has provided experimental data for the clinical application of ischemia therapy in treating pancreatic cancer.

目的 为缺血疗法在胰腺癌的临床应用提供基础研究资料。方法 将人胰腺癌细胞株PC- 3 移植于裸小鼠胰腺右叶,获得小鼠胰腺移植癌,并结扎荷癌裸鼠胃十二指肠动脉、胰十二指肠下动脉及背胰动脉诱导胰腺右叶区域性缺血,观察缺血对移植癌增殖动力学及病理形态学的影响。结果 缺血组移植癌生长缓慢,倍增时间延长,移植癌体积、癌细胞增殖指数及蛋白质含量术后3、7、14天均显著低于对照组(P< 0.01)。光镜下缺血组移植癌呈大片凝固坏死,并可见渐进性坏死细胞、炎症细胞浸润及纤维组织增生;癌周胰腺组织腺泡萎缩,出现纤维化及淋巴细胞浸润。结论 缺血对裸小鼠胰腺移植癌具有确切的杀伤和抑制作用,该模型的建立为临床中晚期胰腺癌的缺血治疗提供了一定的理论基础及实验依据。

Effects of regional ischemia on the growth of transplanted cancer and the pathomorphology of the transplanted cancer and pericancerous tissue were investigated. [WT5”HZ]Results [WT5”BZ]The transplanted cancer grew slower and its doubling time was longer in the ischemic group than in the control. On the 3rd, 7th and 14th day after operation, the size of transplanted cancer, the proliferative index and protein content of the cancer cells were significantly lower in the ischemic group than in the control ( P <0.01). Optical microscopy revealed large areas of coagulation necrosis, necrobiotic cells and the infiltration of inflammatory cells. The atrophy of acini, fibrosis and the infiltration of lymphocyte cells were found in pericancerous tissue. [WT5”HZ]Conclusion [WT5”BZ]Regional ischemia can destroy and inhibit the pancreatic transplanted cancer in nude mice effectively. The ischemia changes of pericancerous tissue may be unfavourable for the growth of the pancreatic transplanted cancer.

目的 为缺血疗法在胰腺癌的临床应用提供基础研究资料。方法 在建立人胰腺癌细胞株裸小鼠胰腺原位移植瘤模型的基础上 ,结扎荷瘤裸鼠胃十二指肠动脉、胰十二指肠下动脉及背胰动脉 ,诱导胰腺右叶区域性缺血 ,观察缺血对裸小鼠胰腺移植癌生长、移植癌及癌周胰腺组织病理形态学的影响。结果 缺血组移植癌生长缓慢 ,倍增时间延长 ,移植癌体积、癌细胞增殖指数及蛋白含量在术后第 3、7、14天均显著低于对照组 (P <0 .0 1)。光镜下缺血组移植癌呈大片凝固坏死 ,周围有渐进性坏死细胞及炎症细胞浸润 ;癌周胰腺组织腺泡萎缩破坏 ,可见广泛纤维化及淋巴细胞浸润。结论 区域性缺血对裸小鼠胰腺移植癌有确切的杀伤和抑制效应 ,癌周胰腺组织的缺血性改变可能不利于移植癌的生长。
