Qinghaosu (Artemisine) suspension in oil was injected im to 30 rhesus monkeys qd×14 d. Damages to several organs and tissues were noticed in 96 and 192 mg/kg/d groups on d 3 after the last injection. They were mainly erythroblastopenia, myeloblastope-nia and megakaryocytosis in bone marrow, degeneration and necrosis of myocardium, degeneration of hepatic cells and epithelia of proximal convoluted tubule of the kidney, and soft tissue injury at the injection site. Qinghaosu (Artemisine) suspension in oil was injected im to 30 rhesus monkeys qd×14 d. Damages to several organs and tissues were noticed in 96 and 192 mg/kg/d groups on d 3 after the last injection. They were mainly erythroblastopenia, myeloblastope-nia and megakaryocytosis in bone marrow, degeneration and necrosis of myocardium, degeneration of hepatic cells and epithelia of proximal convoluted tubule of the kidney, and soft tissue injury at the injection site. Bone marrow and myocardial damages were more prominent than those in other organs. Only mild lesions or no damage could be found on d 35 after the last injection. Therefor, it is concluded that qinghaosu suspension in oil in doses over 96 mg/kg/d might induce transient organ or tissue damages which disappear within 5 weeks. 3 of 4 rhesus monkeys in 192 mg/kg/d dose group died of severe myocardial damage within 1-3 days after the last injection.

恒河猴30只,经连续14天肌注青蒿素油悬剂,于停药后3天,主要在每天肌注96和192mg/kg组中引起多种脏器组织的损伤。表现为:骨髓红系和粒系细胞数减少,成熟发育障碍,巨核细胞增生,心肌细胞变性和灶性坏死(以超微结构病变为主);肝、肾营养不良性改变;淋巴组织萎缩;注射部位的损伤等,其中以骨髓和心肌损伤较为明显。于停药后35天,上述病变明显减轻或消失,表明青蒿素的毒副作用是可逆性的。每天肌注青蒿素24和48mg/kg为轻微中毒剂量;96mg/kg为严重中毒剂量;192mg/kg为致死剂量。每天肌注192mg/kg组的4只猴,有3只于末次给药后1~3天死于严重心肌损伤。

Six anesthetized dogs were bled until their blood pressure dropped to 40mmHg. Serum CPK was then determined every one hour and the values were studied comparatively with the corresponding histological changes. The results showed blood pressure remained at 40mmHg after one hour, serum CPK became obviously higher than that before bleeding,with P<0.05.In two dogs to which blood transfusion was given, the myocardium did not show any obvious changes with HE stain, ing but scattered positively stained areas were seen,...

Six anesthetized dogs were bled until their blood pressure dropped to 40mmHg. As hypotension persisted for 4-6 hours, not only the positively stained areas of myocardial changes enlarged in the allochromatic acid 2R and HBFP stains, but also granular degeneration of muscular fascicles and. sarcoplasmic aggregation appeared in all these three stains. These results show that serum CKP activity changes are positively correlated with the changes of myocardiaL damage.

本实验在用6只犬复制失血性休克模型的基础上,定时采血测定血清肌酸磷酸激酶(CPK)活性值,并同时观察心肌组织学变化。本研究证明,血清CPK活性值变化与心肌组织学改变正相关,因此,观察CPK活性值的动态变化,对休克时急性心肌衰竭的早期诊断有一定价值。

Rat experiments were performed to describe the time course of lipid peroxida- tion(LPO) with various influence upon the heart.The contents of malondialde- hyde(MDA) and the activity level of super- oxide dismutase(SOD)were measured.In addition,the ability of two antioxidants against free radical mediated myocardial damage was evaluated.The experimental groups consisted of rats with intact hearts. rats treated in anterior descendens ramus ligation with ischemia through 45,90, inures,3 hours and 3 days.The treated rats showed characteristic alteration in biochemical parameters and the MAD level and SOD were measured.It was found that the MDA grandually increased and reached its peak value at the end of a 3-hour ligation,whereas the SOD levels decreased sharply between 45 minutes and 3 hours after ischemic insult,It was worth notice that both antioxidants had effects on the suppression of the activation of LPO.

本实验采用结扎大鼠冠脉左前降支法造成心肌梗塞(MI)的动物模型,旨在观察 MI 急性期同一大鼠梗塞区和非梗塞区氧自由基引起的膜脂质过氧化反应(LPO)的时相变化。LPO 的激活程度以 LPO 的产物丙二醛(MDA)的含量及抗氧化酶超氧化物岐化酶(SOD)的活性变化作为观察指标。取材时间分别为手术后0.75、1.50,3.0、72.0小时。并在3小时实验动物组分别给予天然植物抗氧化剂 Z和 R,观察它们对 LPO 的抑制作用。实验结果表明,心肌梗塞区 MDA 呈双向时相变化,手术后3小时达到高峰。SOD 呈下降趋势。在0.75~3小时 MDA 含量升高与SOD 活性下降呈高度负相关。同时观察到所给予的天然植物抗氧化剂 Z 和 R 对 LPO有较好的抑制作用。
