To explore the role of p53 on the progress of cell carcinogenesis,the wild type or mutant-type p53 gene has been transfected into human gastric epithelial cell line-GES-1.Further investigation revealed that wt-p53 changed the morphogical features of cells-some cytoplasmic vaculoes as the sign of cell differentiation were found in the transfected cells and tbe vacuoles were surrounded by microtubulin in wt-p53 transfected cell, and that transfection of p53 gene altered the cell growth behavior-the colony...

To explore the role of p53 on the progress of cell carcinogenesis,the wild type or mutant-type p53 gene has been transfected into human gastric epithelial cell line-GES-1.Further investigation revealed that wt-p53 changed the morphogical features of cells-some cytoplasmic vaculoes as the sign of cell differentiation were found in the transfected cells and tbe vacuoles were surrounded by microtubulin in wt-p53 transfected cell, and that transfection of p53 gene altered the cell growth behavior-the colony formation rate of wt-p53 transfected cells were less than that of wt-p53 and neo vector transfectants.In addition,mt-p53 transfected cells got the anchorage independent growth ability.Tha results show that wt-p53 confers a selective growth advantage on transfected cells and hence leads to malignancy progression.

Objective To compare the growth behavior of epithelium and fibroblast from ameloblastoma and keratocyst and to discuss the recurrence reason for the keratocyst.Methods Epithelial cells and fibroblast from ameloblastoma, keratocyst, dentigerous cyst, radicular cyst and oral mucosa were cultured and observed by phase contrast microscope. Results The epithelial cells of ameloblastoma grew faster, with an irregular area around the cell mass and some small satellite shaped cell mass. The epithelial cells of all the cysts and mucosa grew slower with the cell masses surrounded by fibroblasts which grew in whirl manner with a round and smooth periphery. Conclusion The cells from the ameloblastoma grow faster and more active,and those from the keratocysts grow like the other cysts and the normal mucosa. It is speculated that the ameloblastoma grows in an aggressive manner and it is possible that the recurrence of the keratocyst isn't primarily resulted from its growth behavior as a tumor.

目的观察、比较成釉细胞瘤和角化囊肿细胞的生长特点,探讨角化囊肿复发的原因。方法原代体外培养成釉细胞瘤细胞、牙源性角化囊肿、根端囊肿及口腔粘膜的上皮细胞和成纤维细胞,倒置光相差显微镜观察。结果成釉细胞瘤上皮细胞生长较快,细胞生长晕周围不规则,大细胞生长晕周围有许多卫星样小生长晕,成纤维细胞呈网格状。角化囊肿与其他囊肿和正常粘膜的上皮细胞相同,生长缓慢,细胞生长晕周围规则,由呈漩涡状生长的成纤维细胞围绕。结论成釉细胞瘤细胞生长活跃,而角化囊肿细胞的生长特点与其他囊肿和粘膜相似,提示成釉细胞瘤细胞的浸润性生长特点,推测角化囊肿复发的主要因素不是因为其具有肿瘤的生长特点。

Objective To observe the ordered growth behavior of human urinary bladder cancer cell line under culture in vitro . Methods The suspension of human urinary bladder cancer cells was inoculated to the localized areas. After the cells grew alone the wall to form colonies, macroscopic and microscopic observations of the spreading diameter, spreading rate, cell shape, average cell density, average cell size and dehydrogenase activity and its sensitivity to pH were dynamically performed. Results Under culture conditions, obvious laminar structure appeared in the locally growing colonies of human urinary bladder cancer cells and there was difference of the shape, size, density and dehydrogenase activity and its sensitivity to pH among the cells in different zones. Conclusion Space closing and biodissipation lead to the self organization of the human urinary bladder cancer cells. Consequently they show ordered growth behavior. In this paper, a simple and controlled model of wavy growth of human cells is provided.

目的 观察人膀胱癌细胞 (BIU)在体外培养条件下的生长有序行为。方法 将BIU细胞悬液进行局限区域接种 ,待其形成贴壁生长的群落后进行宏观和显微动态观察及参数测量 (扩展直径、扩展速率、细胞形态、细胞平均密度、细胞平均大小、脱氢酶活性、对pH的敏感性等 )。结果 体外细胞培养条件下 ,局限区域生长的BIU细胞群落能出现明显的层次特性 ,不同区域细胞的形态、大小、密度、脱氢酶活性以及对pH的敏感性均存在差异。结论 空间封闭和生物耗散导致细胞自组织 ,其结果表现为生长有序行为。本实验为人体细胞的波动生长提供了一个简单可控的初步模型
