This paper reported the synergic action of EBV and TPA on the NP epithelial cdlls using the DNA-mediated gene transfer method and the molecular hybridization technique in situ. The experimental results showed that there -was three events in the expression for EBV in vitro (1) EBV gene fragments could incorporate into the NP epithial cells and inserted into host cell DNA. This paper reported the synergic action of EBV and TPA on the NP epithelial cdlls using the DNA-mediated gene transfer method and the molecular hybridization technique in situ. The experimental results showed that there -was three events in the expression for EBV in vitro (1) EBV gene fragments could incorporate into the NP epithial cells and inserted into host cell DNA. So the gene transfer did not need the existence of EBV particles or cell fusion; (2) TPA could promoted the incorporation and the insertion of EBV gene into host cell in vitro; (3) TPA also promoted gene expressions of EBV including a large amount of EBV-mRNA as well as EBNA, a specific gene product of EBV. This study provided an experimental model for gene transfer which reflected the synergetic role on transformation. The ability of promotion of TPA and the integration of EBV gene have been discussed.

Using methods of nucleic acid lybridization In Situ and ultrasonicsplitted gene fragment, this paper reported detections of EBV gene fragments in tumor cells of NPC and Burkitt's lymphoma and host cells transfrred by EBV gene and anal-ysis of gene expression.The experimental results showed that there were three events In Vitro (1) During the latent existence the EBV-RNA transcription and EBNA expression could be demonstrated by molecular lybridazation In Situ; (2) After the EBV gene transfer, the homogenous base squences of EBV-DNA “W” fragment (Bam HI W/Sau Ⅲ a) had inserted in and localized on host cell DNA; (3) The study also found the closed correlation among the EBV-RNA transcription, the EBNA expression and the transformed cell phenotype by cellular agglutination.This study provided a new thought and some approaches for the mechanism on malignant transformation of host cell. It is worthy of further study whether the EBV-DNA “W” segment is the transforming fragment or not.

本文应用原位核酸分子杂交定位和超声切割基因片段技术,对鼻咽癌、Burkitt淋巴瘤细胞和EBV基因转移细胞进行基因片段探测,以及基因表达状态的分析。实验结果表明:(1)EBV基因的潜伏(隐匿)和再激活开放过程中,EBV-RNA的转录和EBNA的表达相关性,可以应用原位核酸分子杂交法显示和定位,(2)EBV基因转移后,与EBV-W片段的同源顺序,已插入和定位于细胞DNA中;(3)EBV基因转移,EBV-RNA的出现与EBNA产生相关;EBNA的表达,又与细胞转化表型——细胞凝集相关。 EBV基因片段的转移,EBV-RNA的转录、基因产物-EBNA的表达以及细胞凝集转化表型的出现,这一检测系统对细胞恶性转化机制提供了新的技术途径和思路。 EBV-“W”片段(BamHI W/Sau IIIa)作为转化片段的设想值得进一步研究。

pSV2-neo gene was transfered into murine hybrjdoma cells ( B889 ) using electric field-mediated gene transfer ( electroporation ). High efficiency of transfer and stable gene expression were detected in these cells subjected to electroporation. Electrical parameters affecting gene transfer, such as the field strength ( Voltage ), pulse durations and pulse frequency ect. and optimum transfer conditions in murine hybridoma cells were studied.

本文应用电脉冲介导基因转移法,获得pSV2-neo基因在小鼠杂交瘤细胞(B889)中的高效转移和稳定表达。研究了电场强度、脉冲宽度、脉冲次数等电场参数对基因转化频率的影响和小鼠杂交瘤细胞最适电转移条件。
