Although only 3 % of the total DNA sequence have been analyzed, the study of the functional genome is already on the agenda and the post genomic era is already with us. The aim of the new era, the era of the proteomics, is to elucidate the expression, structure and function of all the proteins which are after all responsible for life's activities. The main techniques for the separation and identification of all the expressed proteins are two dimentional gel electrophoresis, two dimentional HPLC, mass spectrometry and bioinformatics. Proteome research is not only the major task in life sciences facing us in the 21st century, but also has far reaching implications in innovations for medicine, agriculture and industrial production.

1990年国际上开始了人类基因组研究。尽管目前只解出了约3%的序列,但功能基因组的研究已经开始,后基因组的时代已经到来。新时代的最终目的,是阐明基因组所表达的真正执行生命活动的全部蛋白质的表达规律和生物功能,也即蛋白质组研究。当前主要的研究手段为双向凝胶电泳、“双向”高效柱层析、质谱技术和生物信息学。蛋白质组研究,不仅是21世纪整体细胞生物学新的最重要的内容,而且将为医药、农业和工业的革新提供崭新的思路。

The current methods employing MALDI/TOF/MS and tandem mass spectrometry for peptide-mass fingerprinting (PMF) and peptide sequencing are also discussed. 蛋白质组学是在后基因组时代出现的一个新兴的研究领域,它的主要任务是识别鉴定细胞、组织或机体的全部蛋白质,并分析蛋白质的功能及其模式。因此,揭示蛋白质组中蛋白质间的相互作用关系也是蛋白质组学的重要内容之一。酵母双杂交技术是用来检测蛋白质间是否相互作用的一个非常有效的手段。该技术在酵母蛋白质组研究中的初步成功应用,表明它有望在人类蛋白质组研究中发挥重要作用。
