Nine tea cultivars were employed in the experiment.The plantlets were produced only in the tea“Fuyun No.7”.The unopened flower buds were pre-treated at 5℃ for 48 hours.The developmental stage of the pollens in the anther for inoculation was at the middleanaphase period.The calli were induced on the N_6 medium,supplemented with kinetin(2mg/l),2,4-D (0.5mg/l),L-glutamine(800mg/l)and serine(100mg/l).In order to differentiate shoots the N_6 basal medium was added to zeatin(2mg/l), adenine(20mg/l)and lactalbumin...

试验采用9个茶树品种,但仅在福云7号的花药培养中诱导形成植株。接种花药的小孢子发育期为单核中晚期。供试花蕾经5℃处理48小时。诱导愈伤组织用 N_6基本培养基附加激动素(KT)2.0mg/l,2,4-D 0.5mg/l,谷氨酰胺800mg/l,丝氨酸100mg/l。分化培养基为 N_6附加玉米素(Ze)2.0mg/l,腺嘌呤20.0mg/l 和水解乳蛋白10.0mg/l。形成芽苗后转入含有0.1mg/l IAA 的 N_6培养基诱导长根。愈伤组织诱导在黑暗条件下进行,培养温度为20~25℃。器官分化培养转移到光照与黑暗各12小时的条件下。小苗已成功地移植于田间。其根尖染色体检查结果有单倍体和亚倍体,但未见二倍体植株。还对单倍体植株和亚倍体植株的植物学形态特征进行初步观察。
