A quantitative procedure employing filter paper-partition chromatography is described for the determination of ascorbic acid in plant and animal tissues, using a mixture of n-butanol and 4% aqueous trichloroacetic acid solution as solvent (4:6, v/v). Our experimental results clearly show that the slow breakdown of ascorbic acid during its separation on a paper partition chromatogram is mainly due to the presence of oxygen and such destruction may be completely prevented upon removal of the latter. A quantitative procedure employing filter paper-partition chromatography is described for the determination of ascorbic acid in plant and animal tissues, using a mixture of n-butanol and 4% aqueous trichloroacetic acid solution as solvent (4:6, v/v). The chromatogram is developed in an atmosphere of carbon dioxide with a few per cent of hydrogen sulfide. The average recovery of added ascorbic acid is about 98%. Due to the irregularity in the percentage reduction of dehydroascorbic acid in the chromatographic chamber in the presence of H_2S, the procedure can only be used to determine the total content of ascorbic acid after preliminary reduction of the dehydro-compound.

The effect of sodium pentobarbital in mice(sleeping time) and rabbits (EEG)at different periods after a single total-body irradiation at a dose of 500r or1000r was investigated. The effect of sodium pentobarbital in mice(sleeping time) and rabbits (EEG)at different periods after a single total-body irradiation at a dose of 500r or1000r was investigated. The rate of metabolism of pentobarbital in these irradiated animals was also determined. At the early and the recovery stages (1—5 days and 16—30 days after 500r X-irradiation), the sodium pentobarbital (60mg/kg i. p.)sleeping time in mice was not affected. However, at. the height of radiation sickness, i. e. 5—15 days after irradiation, the sleeping time was significantly prolonged. Administration of sodium pentobarbital (20mg/kg i. v.)to rabbits with severe radiation injury caused by exposing to a single total-body dose of 1000r of Υ-irradiation (Co~60), produced a deep depressing effect as indicated by changes in the EEG, while prior to irradiation the same dose of pentobarbital caused only negligible depression. The rate of disappearance of the injected pentobarbital from X-irradiated skinned whole mice was significantly slower than that of the control animals. This observation was substantiated by the rate of biotransformation of the drug in vitro by liver slices from X-irradiated mice. When a comparison was made between the concentrations of the drugs in irradiated and control mice at the wake-up time, it was found that the irradiated animals woke up at a total body level significantly lower than the controls. It would appear that the irradiated animals were more sensitive to the drug. These results would suggest that both the decrease in the rate of metabolism of the drug and the increase of the sensitivity of the central nervous system to the drug contributed to the change of the response of the irradiated animals to pentobarbital.

小鼠照射500伦琴X线后,在照射后初期对戊巴比妥纳的反应无明显改变,但在病熾期則反应显著加強,及到恢复期反应又渐恢复正常。在放射病病熾期,戊巴比妥鈉在小鼠体内的消失减慢,肝切片对其生物转化速度也相应减慢。若給小鼠注射戊巴比妥鈉入睡后,待其刚醒时,测定体内药物含量,則照射组明显低于对照組。当家兔照射1000伦琴γ线后的病熾期,給予照前仅引起正常兔脑电图轻度抑制的戊巴比妥鈉的剂量时,可使脑电图卽口刻呈现深度抑制状态。由这些結果看来,受照射机体对戊巴比妥鈉反应性增加的原因,一为受照射机体对該葯的转化能力减低,一为在放射病病熾期,动物的中枢神经系统处于抑制占优势状态。因而对戊巴比妥作用的敏感性增強。
