Objective To obtain new compounds with the antitumour activity from a secondary metabolite of marine microorganisms. Methods 200 strains of marine microorganisms which were isolated from the Yellow Sea, the Bohai Sea and the offing of Liaoning Province of China have been screened by the model of inducing morphological abnormalities of Pyricularia oryzae P-2b. Results 6 strains of microorganisms with a better bioactive substance in their fermented broth were obtained (Staphylococus delphini, Paecilomyces sp.,...

Objective To obtain new compounds with the antitumour activity from a secondary metabolite of marine microorganisms. Methods 200 strains of marine microorganisms which were isolated from the Yellow Sea, the Bohai Sea and the offing of Liaoning Province of China have been screened by the model of inducing morphological abnormalities of Pyricularia oryzae P-2b. Results 6 strains of microorganisms with a better bioactive substance in their fermented broth were obtained (Staphylococus delphini, Paecilomyces sp., Aspergillus terreus, Penicillium sp., Sinopaecilomyces, Bacillus marinus);Four of the strains Paecilomyces sp., Penicillium sp., Sinopaecilomyces, Bacillus marinus exhibited better stability of inheritance and growth. Conclusions Their bioactive substances had better stability of heating. All of the above results will be helpful for the separation of antitumour bioactive compounds.

目的从海洋微生物的次级代谢产物中发现新的具有抗肿瘤活性的新化合物以用于新的抗肿瘤药物的研制开发。方法以稻瘟霉孢子形态变异为指标的生物活性物质筛选模型 ,对 2 0 0株海洋微生物资源菌株进行了反复筛选。结果得到了 6株在其发酵液中能够产生较强生物活性物质的菌株 ,即海豚葡萄球菌 (Staphylococusdelphini)、拟青霉 (Paecilomycessp )、土曲霉 (Aspergillusterreus)、青霉 (Penicilliumsp .)、中华拟青霉 (Sinopaecilomyces)、海洋芽孢杆菌 (Bacillusmarinus) ;并且对这 6株菌株的生长发育稳定性 ,遗传稳定性和生物活性物质的热稳定性进行了考察 ,发现其中的 4株菌株 ,即拟青霉 (Paecilomycessp )、青霉 (Penicilliumsp )、中华拟青霉 (Sinopae cilomyces)、海洋芽孢杆菌 (Bacillusmarinus)的各项稳定性良好。 结论为具有抗肿瘤活性物质的追踪分离奠定了基础

The physical and chemical stability of the bioactive compound obtained from Streptomyces 702's fermented broth were tested. The bioactive compound from fermented broth has antibacterial action on B.subtilis, Saccharomyces cerevisiae and Penicillium citrinum. The bioactive compound is ultraviolet-stable, acid- base-stable under pH3.0-pH12.0 and heat-stable treated by 100℃ for 10h, and even by 121℃ for 60min.The statistics will be useful for the further study on the bioactive compound.

通过对链霉菌702所产生物活性物质的稳定性测定研究,试验结果表明,链霉菌702发酵液分别在100℃和121℃处理10h和1h,紫外线照射3min和在pH3~pH12缓冲液中处理60min后,分别对枯草芽孢杆菌、啤酒酵母和青霉的抑菌活性不减。说明该抑菌物质对热和紫外线稳定,在pH3-pH12条件稳定。本研究为进一步对该抑菌活性物质分离提取工艺和其在实践上的应用,提供了有益的试验数据。

The inhibitory action of the bioactive compound produced by Streptomyces 702 ,Nisin and Natamycin on Aspergillus niger,Penicillium chrysogenum,Trichoderma viride,Bacillus subtilis,Bacillus cereus,Bacillus thuringensis,Candida utilis and Saccharomyces cerevisiae was tested. The L_(C50) and the L_(T50) of the bioactive compound produced by Streptomyces 702,Nisin and Natamycin on the spore of bacteria and fungus were obtained. The result showed that the inhibit effect of the bioactive compound produced by Streptomyces 702 was more powerful than those of Nisin’s and Natamycin’s. The experimental results are useful for the application study of Streptomyces 702.

通过对链霉菌702所产生物活性物质和纳他霉素及Nisin分别对黑曲霉、青霉、绿色木霉、枯草芽孢杆菌、蜡质芽孢杆菌、苏云金芽孢杆菌、产朊假丝酵母、啤酒酵母的抑制作用研究,试验结果得出链霉菌702所产生物活性物质对各种指示菌的半致死浓度和半致死时间,证明链霉菌702所产生物活性物质抑制效果比纳他霉素及Nisin强。试验结果为链霉菌702所产活性物质应用提供了有益的试验数据。
