Surface display is a newly developed gene manipulation technology by which fragments of foreign peptides are expressed as fusion proteins displayed on the phage or cell surface, keeping their spatial structure and biological activity relatively independent. This technology may be used for studying the properties, mutual recognition and interaction of peptides (proteins), so as to survey very large display libraries for target functional peptides. There are three display systems, including filamentous bacteriophage, T 4 and T 7 phage,and cell surface systems, mosthy used so far. Display librariesinclude recombinant phage antibody libraries, random short peptide libraries, peptide comformation libraries, cDNA display libraries, and gene mutant display libraries. The surface display technology can be used for the production of artificial antibody and vaccine, the determination of antigen epitopes and protein interactive sites, the isolation of regulatory molecules, the study of cell surface engineering, the development of peptide drugs, and the experimentally directed evolution of biomelecules.

The gene Ⅷ of Filamentous bacteriophage was cloned to pKK 223-3 vector. A oligonucleotide encoding the residue of six amino acid was inserted to the above modified plasmid and the hybrid phage was made. SDS-PAGE showed that the foreign peptide gene worked normally and proteins of the gene were displayed on the major coat protein of Filamentous bacteriophage.

The synthesized random oligonucleotides which encode nonapeptides are cloned into the filamentous bacteriophage vector fUSE5,and transformed into the competent E.coli MC1061 by multielectroporation,10 10 clones are obtained in which each peptide expressed as a fusion to the minor coat protein pⅢ and displayed on the surface of the phage.Affinity selection has been used to identify potent ligands for the human type Ⅳ collagenase and a family of collagenase binding bacteriophage ligands was obtained.ELISA results showed 20 positive clones that could bind the type Ⅳ collagenase specifically.Sequences showed two relatively short conserved subsequences:WDXXD and WVGXXR,the sequence WDXXD is homologous to the ScFv variable region of the type Ⅳ collagenase.It is showed that the peptide library is a powerful tool for the selection of the potent peptide ligands for proteins.The construction and screening of the nonapeptide library forms the bases for the further application.

将人工合成的编码九肽的随机序列DNA片段克隆进丝状噬菌体表达载体fUSE5,经多次电击转化和表达,获得肽段与噬菌体pⅢ蛋白融合并展示在噬菌体表面的随机序列九肽表位肽库。库容量达1010个克隆。以Ⅳ型胶原酶为靶蛋白,采用亲和纯化筛选模式,从中筛选出Ⅳ型胶原酶结合肽。进一步ELISA检测筛选出与Ⅳ型胶原酶特异结合的20个阳性克隆。序列分析发现一组肽含有WDXXD的共同序列,一组含有WVGXXR的共同序列。其中WDXXD的序列与Ⅳ型胶原酶单链抗体可变区序列同源。结果表明,多肽库是筛选蛋白特异结合肽的有力工具,表位九肽库的构建和筛选方法的建立为进一步应用筛选具有高亲和力的特异结合肽奠定了基础。
