Isolated pollen protoplasts of Nicotiana tabacum N364 K + m were fused with mesophyll protoplasts of Nicotiana rustica using PEG-high Ca/pH method. The cells resulted form fustion between immature (early-middle bicellular) pollen protoplast and mesophyll protoplast could divide to produce multicellular structures and microcalli when being cultured in a selection KM 8p ,medium containing 50μg/ml kanamycin. The cells derived from fusion of mature pollen protoplast with mesophyll Protoplastant only...

Isolated pollen protoplasts of Nicotiana tabacum N364 K + m were fused with mesophyll protoplasts of Nicotiana rustica using PEG-high Ca/pH method. The cells resulted form fustion between immature (early-middle bicellular) pollen protoplast and mesophyll protoplast could divide to produce multicellular structures and microcalli when being cultured in a selection KM 8p ,medium containing 50μg/ml kanamycin. The cells derived from fusion of mature pollen protoplast with mesophyll Protoplastant only formed tube-like structures instead of cell division. The results of the experiment indicated that the developmental stage of pollen protoplasts involved in fusion played an improtant role in deciding the developmental pathways of the fusion products: The mature pollen protoplast when fused with somatic one exhibited strong influence of gametophytic deveolpmental trends towards tube growth, whereas the younger pollen protoplast had some potential of sporophytic development which did not negatively affect division ability of its somatic partner. Regeneration and genetic characterization of plants from the microcalli are underway.

In order to investigate the DNA polymorphism of Cladosporium carrionii and Cladophialophora ajelloi, and to find the relationship of DNA patterns with morphology and geographic origin, the total DNA was extracted by using benzyl bromide method, and random amplification of polymorphic DNA(RAPD) assay was used to the study of genetic similarity(GS) of 21 isolates from five countries(19 of Cladosporium carrionii, 2 of Cladophialophora ajelloi). Results:(1)Of ten random primers used, three gave good reaction.(2) The DNA fragment patterns of 19 isolates of Cladosporium carrionii showed high genetic similarity, but a little difference in comparison with that of 2 isolates of Cladophialophora ajelloi. The RAPD patterns of two isolates showed high genetic similarity with most Chinese isolates of Cladosporium carrionii(GS∶85%).(3) The genetic similarity of Cladosporium carrionii are closely related to the geographic origin, the DNA patterns of isolates from the same country showed high similarity and located in the same group of dendrogram. RAPD assay can be used for the study of epidemiology and genetic characterization as a simple, rapid method.

研究卡氏枝孢霉与阿耶罗枝孢瓶霉的DNA多态性,了解两菌在DNA型别与形态学及菌种来源地域的相互关系。采用溴苯提取法提取DNA,以随机扩增DNA多态性方法对来自五个国家的21株分离株(19株卡氏枝孢霉,2株阿耶罗枝孢瓶霉)进行研究。结果:①10个随机引物筛选出3个具有较好DNA扩增的引物。②19株卡氏枝孢霉的DNA带型具有较大的遗传相似性,阿耶罗枝孢瓶霉的DNA带型与卡氏枝孢霉略有不同,但与中国株具有较大相似性(85%的遗传相似性)。③卡氏枝孢霉的遗传相似性与菌株的来源地域关系密切,同一国家的菌株具有相似的DNA带型,位于树状图的同一树组中。结论:随机扩增DNA多态性研究发现卡氏枝孢霉具有一定的种内差异,其DNA带型与菌株的来源地域关系密切,阿耶罗枝孢瓶霉的DNA带型与某些株卡氏枝孢霉更相似。本研究方法简便、快速,可用于流行病学及基因学特征的研究

扩增片段长度多态性(AFLP)是建立在PCR和RFLP技术基础上的一项新的DNA指纹技术,可广泛应用于种系鉴定、遗传多样性检测、群体分类研究、构建遗传图谱、标定目的基因等方面。本文结合AFLP的最新发展,对该项技术的原理、特点、分析流程、优越性进行了概述,提出了AFLP分析中应注意的问题,并探讨了该技术的应用前景。
