Genetic characterization of 156 measles viruses isolated from throat swabs or urine specimens of measles patients using B95a cell line in nineteen provinces of the People's Republic of China during 1995-2002 was studied.The sequence analysis of 450 nucleotides of COOH-terminal of nucleoprotein(N)genes of 156 isolates indicated that all of the Chinese viruses were members of H1 genotype except Hunan95-6 and Henan99-1,that was same with Chinese measles vaccine strain S191,and belonged to genotype A.These Chinese measles viruses of H1 genotype were more diverse as compared to the other genotypes and differed from genotype A(Edmonston)at 6.00%~7.33%,from genotype H2(China94-1)at 5.11%~7.56% and from other 17 genotypes as much as 11.56% in the COOH-terminal of the N gene.The Chinese measles virus H1 genotype is the most genetically divergent within single genotype described so far.The sequencing data suggest that H1 genotype of 154 Chinese measles viruses can further be divided into two small subgroups,the H1a and the H1b,except Shanxi100-6 and Hunan-23.The H1a subgroup differed from H1b by 2.00%~4.00% at the nucleotide level in the COOH-terminal of the N gene.It is helpful to develop measles virus surveillance and set up measles virus strain bank and virus gene bank to accelerate measles control and eliminate measles in future.

Objective To ascertain the genetic characterization and genotype of measles viruses isolated in Shanghai region, in 2005. Methods Measles virus was isolated from throat swab specimens collected from suspected measles cases and 450 bp fragment of C terminus of nucleprotein (N) gene was amplified by RT-PCR. Sequence analysis was conducted to ascertain the genotype and to compare the difference of nucleotide with other measles virus strain published in GenBank. Results 4 measles viruses were isolated from 10 throat swab specimens, and the sequence analysis indicated that they belonged to H1 genotype. The homogeneity of 450 nucleotides in the C terminal of the N gene was at 98%-98.2% as compared to H1 genotype (China93-7) .They differed from genotype H2 (China94-1) at 6.4%-6.9% and from genotype A (Edmonston) at 6.7%-6.9%, from measles vaccine (Shanghai191) at 7.6%-8.0%. They differed from the other measles viral strain isolated in China in 1993-2005 at 0.2%-3.7%. The variation within 4 isolated measles viruses was at 0.7%-1.3%. Conclusion It was H1 genotype measles viruses, which are the native viruses in China that led to the outbreak of measles in Shanghai, in 2005.

目的了解2005年上海部分地区麻疹暴发流行株的基因型别和特性,加强麻疹病毒学监测。方法采集暴发麻疹患者咽拭子标本分离病毒,通过RT-PCR扩增麻疹病毒N基因C末端450 bp核苷酸并进行序列测定,与GenBank中麻疹病毒各基因型参考株及国内其他地区的麻疹分离株进行基因比较。结果从10例麻疹患者的咽拭子标本中共分离麻疹病毒4株,均属H基因组H1基因型,型内变异0.7%-1.3%。与H1型代表株China93-7的基因同源性为98%-98.2%;与H2基因型代表株China94-1的基因差异为6.4%-6.9%。与A型代表株Edmonston的基因差异为6.7%- 6.9%;与中国麻疹疫苗株Shanghai191的基因差异为7.6%-8.0%,与国内其他地区麻疹流行株基因差异为0.2%-3.7%。结论2005年上海部分地区麻疹暴发是由H1基因型病毒引起,为中国本土流行株。

The genetic characterization of vaccine mutant polioviruses isolated from AFP cases in China was analyzed for the purposes of offering scientific basis for the strategy of OPV/IPV administration and maintaining the polio-free status in China, as well as the polio eradication in the world. Based on the results of intratypic differential (ITD) test of all the vaccine-associated polioviruses isolated from AFP cases in 2000-2001, the complete genome of 5 isolates (3 from Guizhou Province, 2 from Shandong Province)...

The genetic characterization of vaccine mutant polioviruses isolated from AFP cases in China was analyzed for the purposes of offering scientific basis for the strategy of OPV/IPV administration and maintaining the polio-free status in China, as well as the polio eradication in the world. Based on the results of intratypic differential (ITD) test of all the vaccine-associated polioviruses isolated from AFP cases in 2000-2001, the complete genome of 5 isolates (3 from Guizhou Province, 2 from Shandong Province) were sequenced to analyze the homology of nucleotide and amino acid sequences.Three isolates from Guizhou Province had completely the same nucleotide sequences, but recombined with SabinⅢ in 3A region (nt5343-5353). Compared with Sabin strains, 19 nucleotide substitutions were found, 10 in typeⅡ region (4 of them in VP1 region)and 9 in typeⅢ region. The homology of nucleotide and amino acid sequences in VP1 region were 99.56% and 99.34% respectively as compared with that of SabinⅡ. Two isolates from Shandong Province had no recombination and shared 16 nucleotide substitutions, seven and four nucleotide substitutions were found in their VP1 region respectively as compared with that of SabinⅡ, and the homology of nucleotide and amino acid sequences were 99.22%-99.56% and 99.0%-99.67% respectively. Nucleotide substitutions were found at two important attenuation sites (nt481 and nt2909) in each of these 5 isolates. These 5 isolates were isolated from two transmission lineages, but they shared 3 nucleotide substitutions (nt481, nt2 909 and nt2 992), which were not in recombination region. The co-action of these 3 substitutions may influence viral transmissibility. No evidence indicated that intertypic recombination of vaccine mutant polioviruses may enhance the viral neurovirulence and transmissibility.
