The effects of the combined use of two kinds of cytokines in both clinical and experimental studies reported in the literatures were reviewed. A number of laboratory studies have recently demonstrated that any one of IL-3, IL-6, IL-11 used in combination with any one of G-CSF, GM-CSF, Tpo or SCF all exhibited synergetic effect stimulating hematopoiesis in mice and primates. However, clinical effects of combined administration of IL-3 with GM-CSF or IL-6 with G-CSF in patients with MDS, advanced or high grade...

The effects of the combined use of two kinds of cytokines in both clinical and experimental studies reported in the literatures were reviewed. A number of laboratory studies have recently demonstrated that any one of IL-3, IL-6, IL-11 used in combination with any one of G-CSF, GM-CSF, Tpo or SCF all exhibited synergetic effect stimulating hematopoiesis in mice and primates. However, clinical effects of combined administration of IL-3 with GM-CSF or IL-6 with G-CSF in patients with MDS, advanced or high grade sarcoma and in children with recurrent/refractory solid tumors were no better than that of single administration. Otherwise, the toxicity in combined administration is significant. Extensive studies on indication and dosage for combined administration of these cytokines should be necessary in order to obtain good effects on hematopoietic recovery in human being as additive or synergetic effect.

本文综述了两种细胞因子联合应用效果的实验和临床研究进展。在小鼠和猴IL-3,IL-6和IL-11与G-CSF,GM-CSF,SCF或Tpo联用促进造血恢复的效果比单因子好,而对晚期肿瘤或高恶性肉瘤的病人以及实体瘤的儿童来说,IL-3+GM-CSF和IL-6+G-CSF联用的临床效果尚不尽人意,且毒副作用明显。因此,要获得较理想的联合应用效果,还需要对联用时的适应证和剂量等因素给予慎密的研究。
