    The conformation of the tetrapeptides Asn-Ala-Gly-Ala (NAGA) and Asn-D-Ala-Gly-Ala-NH_2(NDAGA) and the pentapeptide Asn-Ala-Gly-Ala-Asn (NAGAN) in solution were studied using the circular dichroism method. All the peptides were chemically synthesized employing the liquid phase method and were chromatographically pure. At low pH the far UV CD spectra of aqueous solutions of NAGAN show a positive extremum at 215nm and a negative extremum at 232.5nm. The curve of[θ]_(215) versus pH is similar to a titration curve with a midpoint at pH2.5~3.0. Accordingly, the molecule of NAGAN may assume a β-turn structure under these conditions. In neutral or alkaline aqueous solutions, NAGAN shows spectra with a single negative extremum at 196nm. It can be deduced that the molecule now assumes a random coil form. In solutions in 90% ethanol (EtOH) or trifluoroethanol (TFE) at pH5.8, NAGAN reveals CD spectra with a negative 218nm extremum. The [θ]_(218) values are independent of the concentration of NAGAN. An intramolecular β-pleated sheet-like structure (Fig 9) was suggested to explain the negative peak.The CD spectra of NAGA show a single positive band at around 215nm. The possibility of NAGA forming a β-turn was discussed. Only the 196nm negative extremum can be seen in the spectra of NDAGA. It seems that NDAGA is in the form of a random coil.Pang et al. reported that all these peptides possess analgesic activity. It seems therefore that the analgesic activity is not related to the special conformation of the peptides. It is only the chemical structure that matters.
