The object of this paper is to discuss the protective effects of taijitongtian solution on ischemic brain injury, the model of ischemic brain was made by ligaturing middle cerebral artery. We observed the changes of the ischemic areas , content of MBP in blood and cerebrospinal fluid and LPO and SOD in blood and brain tissuse of observed group and con-trolied group. All these suggested that taijitongtian solution could significantly protect brain tissuse and abate the ischemic injury.

本文旨在研究太极通天液对脑缺血性损伤的保护作用。用结扎狗的大脑中动脉制造脑缺血模型,观测用药组与对照组之间脑组织缺血区大小,血和脑脊液中髓鞘碱性蛋白的变化,以及血和脑组织中过氧化脂质和超氧化酶的改变。其结果是用药组较对照组有显著性减小缺血区、减少髓鞘碱性蛋白释入脑脊液和血中,并使脑组织中的超氧化酶相对减少较少,从而使过氧化脂质的生成较少。根据实验结果,太极通天液有明显的保护脑组织和减小脑缺血性损伤的作用。

A number of Chinese herbs have been indicated to have protective effects beforecerebral ischemic attacks in man. Polydatin, a glycoside isolated from the roots of Polygonum cuspidatum, is one of the major active ingredients of the Chinese herb. A number of Chinese herbs have been indicated to have protective effects beforecerebral ischemic attacks in man. Polydatin, a glycoside isolated from the roots of Polygonum cuspidatum, is one of the major active ingredients of the Chinese herb. To investigate the effects of polydatin on free radicals in cerbral ischemia, a model of ischemia/reperfusion in rat brain was used. After polydatin administration, the contents of brain water and malondialdehyde (MDA) decreased, and the activities of superox-ide dismutase(SOD), catalase (CAT) and glu-tathione peroxidase (GSH-Px) were elevated to certain extent in cerebral cortex and hippocampus of the rats. Protective action of polydatin in ischemia/reperfusion induced brain injury has been suggested. The effect of polydatin was re-latd to the concentrations of the polydatin administered. The results showed that 12 mg ?kg-1 polydatin (iv) had the most significant effect on free radicals in ischemic brain injury. The results of this study may be useful in designing new drugs in the treatment of strokes and cerebral vascular diseases.

采用大鼠脑缺血再灌注模型,观察系列剂量的白藜芦醇甙(Polydatin,Pol)对大鼠大脑皮层及海马部位的脑含水量、丙二醛、GSH含量及超氧化物歧化酶(Super-oxide dismutase,SOD)、过氧化氢酶(Catalase,CAT)、谷胱苷肽过氧化物酶(Glutathione Peroxidase,GSH-Px)活性的影响.结果表明,Pol能不同程度地降低脑组织的过氧化脂质含量,提高SOD、CAT和GSH-Px活性,降低脑含水量,减轻自由基反应对脑组织的损害,对缺血的脑组织具有保护作用.Pol的作用与所用剂量有明显关系,用Pol 12mg·kg~(-1)(iv)时作用最明显,剂量过高或过低均可影响其对脑组织的保护作用.

OBJECTIVE:To study the content change of endothelin (ET)and cerebral protective action of Ginkgo Biloba extract (EGb761)after cerebral ischemia in rats.METHODS:Animal model was established by middle cerebral artery occlusion(MCAO)in rats and the ET contents in cerebral ischemic area and plasma,the infarction size and edema volume in brain,before and after EGb761 treatment were determined.RESULTS:The ET level and cerebral ischemic area and edema volume were reduced by EGb761 treatment in rats 24 h after MCAO and a highly positive correlated relation between ET concentration and infarction size was obtained(r=0.92, P <0.01).CONCLUSION:ET may be deleterious to cerebral infarction and the protective action of EGb761 in ischemic brain injury may be related to its action in lowering ET concentration.

目的:研究脑缺血后内皮素(ET)变化及银杏叶提取物(EGb761)的脑保护作用。方法:建立大鼠大脑中动脉阻断(MCAO)模型,分别检测脑缺血和EGb761治疗后脑缺血区及血浆中的ET水平、脑梗塞范围和水肿体积。结果:MCAO后24h,EGb761降低了ET浓度,减小了脑梗塞范围和脑水肿体积。缺血脑组织中的ET浓度与梗塞范围之间呈显著正相关(r=0.92,P<0.01)。结论:EGb761对缺血性脑损伤的保护作用可能与其抑制脑缺血急性期ET的生成有关
