The distribution and metabolic fate of the ~(125)I-urokinases modified by soluble polymers in mice were investigated by comparison. Through the investigation, the characteristics and the future application toclinical practice of modified urokinases were discussed. It was shown that the radioactivity of ~(125)I-urokinases modified by soluble dextran sulfate in the blood of mice having been given intravenous injection evidently decreased more slowly than the native ~(125)I-urokinase did. The distribution and metabolic fate of the ~(125)I-urokinases modified by soluble polymers in mice were investigated by comparison. Through the investigation, the characteristics and the future application toclinical practice of modified urokinases were discussed. It was shown that the radioactivity of ~(125)I-urokinases modified by soluble dextran sulfate in the blood of mice having been given intravenous injection evidently decreased more slowly than the native ~(125)I-urokinase did. And the life of modified urokinases in blood was extended.The isotope experiment also showed that the distribution of the radioactivities of nature ~(125)I-urokinase and modified ~(125)I-urokinases increased rapidly in kidney and in liver, and after 24 hours from of injection they were almost disappeared. But in heart, lungs and spleens, the variance were very small from the beginning to the end, and in these tissues there was no storage of radioactivity. The experiment also showed clearly that the variance of ~(125)I-urokinase in liver and kidney was very different from that of modified ~(125)I-urokinases. The distribution of native ~(125)I-urokinase was evidently higher than that of modified ~(125)I-urokinases in kidney. But, in liver, the distribution of native ~(125)I-urokinase was a little bit lower than that of modified ~(125)I-urokinases.It was found that the urokinases modified by soluble dextran anddextran sulfate not only have very good characteristics for fibrinolysis,but also extended the life of the urokinases in blood. Therefore that themodified urokinases may have bright future in the application to cllncalpractice.

本文用放射性同位素~(125)Ⅰ示踪方法对比研究了水溶性大分子修饰酶与天然酶在小白鼠体内的滞留率;通过对标记物在各脏器中的分布及代谢的研究,对修饰酶的性质及临床应用前景做出了判断。 实验结果表明;右旋糖酐及右旋糖酐磺酸酯修饰了的尿激酶明显地延长了它们在血液中的停留时间;尿激酶进入体内后,主要分布于肝及肾脏中,脾、肺及心脏中分布很少;修饰酶与天然酶在肝及肾中的变化很不一致,天然酶在其主要排泄器官肾中分布较高,而修饰酶在肝中的分布则略高于天然酶。
