The recent opportunity of examining the faecal material of the water-scorpion Leccotrephes japonensis revealed a flagellate which had not previously been described from this host. After fuller investigation I found that it is a species new to science and should be referred to the genus Retortamona; I have named it Retortamonas leccotrephae sp. nov.The specimens of Leccotrephes japonensis used for studies on the parasitic flagellate R. leccotrephae, were collected from ponds on the outskirts of Shanghai, China....

After fuller investigation I found that it is a species new to science and should be referred to the genus Retortamona; I have named it Retortamonas leccotrephae sp. nov.The specimens of Leccotrephes japonensis used for studies on the parasitic flagellate R. leccotrephae, were collected from ponds on the outskirts of Shanghai, China. Apart from studying the living flagellates, at the same time stained specimens were prepared for the detailed study of various structures. These specimens were fixed either in Hollande's or in Schaudinn's fluid; stained in Heidenhain's haematoxylin, and destained in 2% aqueous phospho-tungstic acid. Moskowitz' (1950) modification of the Protargol impregnation method was also used for staining.In the living state, the flagellare has a slender, slipper-shaped trunk with an anterior dorsal flexure. The mean body size, exclusive of the posterior spike, is 12.8μ×4.8μ. The length of the slender posterior spike varies from 4.6μ to 15.8μ. There are two flagella of unequal length. The longer one extends forwards and lashes rapidly, causing the animal to rotate and to pursue a special course. The shorter flagellum is directed backwards and usually lies in the cytostome; it moves in an undulating manner.In fixed and stained preparations, the flagellates, not including the posterior spike, show a size-range of 6.2×2.2μto 15.6×5.4μ, with a mean of 11.9×3.4μ. The spike varied from 4.6μ to 13.6μ long, with an average length of 11.2μ. In general, a normal specimen of R. leccotrephae has a slender body; the anterior portion is narrower than the postcerior, and is markedly bent as shown in the living indivi.duals. The posterior portion of the body usually has a dorsal convexity, and the widest region is located near the two-thirds of the body-length from the anterior end.Near that edge of the nuclear membrane lying next to the cytostome, there are two minute, but separate, basal granules which give rise to the two flagella. The anterior flagellum is about as long as the body proper: much longer and more slender than the posterior flagellum, which usually lies within the cytostome, and commonly shows a series of two or three undulations; it often stains more deeply than the anterior flagellum.In hematoxylin preparations the nucleus is well stained, and clearly shows a layer of chromatin granules lying against the nuclear membrane, and a large central endosome, composed of a mass of granules.The cytostome is a large structure in this animal. It usually occupies about twothirds of the total length of the body proper. Along the margin of the cytostome there are two deeply staining fibres, the one on the right margin is longer than that on the left.Both the precystic forms and cysts of R. leccotrephae have been observed. The body of precystic individuals which are preparing to encyst, becomes smailer and rounds up; meanwhile, the chromatin of the nucleus condenses and forms a deeply staining endosome. The anterior flagellum is still disposed freely, and no cyst wall has as yet been formed. The border-line of the cytostome is distinct, but the two basal granules are visible only in favourable preparations. The mature cysts are nearly peanut-shaped, and are about twice as long as wide. In the stained specimens they are about 4.6μ long and 2.4μ wide. The cyst wail is of uniform thickness, and the nucleus is visible at one end. The other conspicuous structures within the cyst are the two flagella and the cytostomial fibres, which are arranged as they are in the trophozoite, except that the anterior flagellum is here directed posteriorly.DISCUSSION & SUMMARYIn its general body-form, R. leccotrephae closely resembles Mackinnon's R. agilis ('11) from crane-fly larvae, and also Corradett's R. gryllotalpae ('37) from the mole cricket, but it is especially like Geiman's R. caudacus ('32) from the aquatic larvae ef certain beetles.

The scientific application of an ideal rotation system on paddy fields,has now been considered as a very important problem for ensuring highergrain yields and promoting soil fertility.As the commencing work onrotation researches,authors had selected to study the Rice-Cotton rotationsystem in Yuyao,Chekiang Province.A series of investigations and ex-periments were conducted in the year of 1955,in order to realize thevariations of soil chemical properties and rice growth on those rotatedpaddy fields.The results obtained are summerized as follows:(1)During the period of Cotton rotation in paddy fields,the organiccontents in the soil tend to decrease,while the total exchangeable bases andavailable phosphorus show an apparant increase.By cultivating rice in suchfields,a reversible tendency appears,namely manifesting an increase ofsoil organic matter content and a decrease of both total exchangeable basesand available phosphorus contents.Besides,the formation of more am-monical nitrogen is found in the later case.(2)The Rice-Cotton rotation will assuredly increase the rate of ricegrowth as well as the yields of grains.Comparing with the unrotatedplot,paddy fields so rotated show an average increase of 9% in grainyeilds,as confirmed by field experiments.(3)In those rotated paddy fields,winter crop-green manure(Medicagodenticulata wild)grew vigorously and showed an increase of 70% in yields(based on fresh weight)as compared with unrotated paddy fields.(4)Rice-Cotton rotation may adjust the rate of the alternation of ac-cumulating and decomposing of soil organic matter,which we know willmarkedly promote soil fertility in paddy fields.Therefore,to save the ap-plication of manure and solve the shortage of fertilizers in grain produc-tion,the use of Rice-Cotton rotation system in certain district,is ofparamount importance at present time.

(1)稻田轮种棉花期间,土壤有机质含量有减低的趋势,但土壤代换性盐基总量及速效磷的合量均增高。轮种水稻期间,土壤有机质又有增加的趋向,而代换性盐基总量及速效磷则渐次减低。此外,土壤中铵态氮则有增多的趋势。(2)稻棉轮种能增强水稻的生长势,提高水稻的产量。根据对比试验及统计资料,一般比连种区增产9%左右。(3)稻田轮种棉花后,冬作绿肥生长良好, 鲜草产量比一般连种稻田增加70%以上。(4)在保证增产的前提下,轮种区的水稻基肥用量,一般可比连作稻区节省40%左右,而轮种区的绿肥产量又能增加,因此可自轮种田内割出60%的绿肥鲜草,作为其他稻田的基肥,而达到全面增产的目的,这在目前肥料普遍不足的情况下有其重要意义。

The apparatus is essentially composed of a ground glass cylinder made from suitable hard glass tubing, which is filled with ice and rotates mechanically in horizontal position in a closely fitting trough of hard porcelain. (1) In the present report a simple type of bacterial mill is described. The chief advantages of this apparatus are high percentage of disintegration, short duration of grinding and repeatability of results. The apparatus is essentially composed of a ground glass cylinder made from suitable hard glass tubing, which is filled with ice and rotates mechanically in horizontal position in a closely fitting trough of hard porcelain. (Fig. 1 and 2). It allows with the help of an abrasive, such as glass powder, to break bacteria (Escherichia coli) to an. extent of 99.99%. The fast and highly efficient breaking of the cells permits the preparation of particulate fractions possessing enzymatic activities, contaminated only with an inconsiderable amount of viable bacteria. Because the grinding process does not require any manual operation besides adding the bacteria abrasive mixture and removing the ground material, extracts of comparatively reproducible activity can be obtained from the same batch of microorganisms. Thus the succinic and malic oxidase activity in extracts from the same lot of bacteria did not differ by more than ±10% and ±16% from the average activities respectively. (2) The extracts obtained from the ground E. coli prepared with our mill were separated by differential centrifugation into a particulate (sedimenting between 5,000-20,000)×g during 2 hours) and a non-sedimentable fraction (supernatant after centrifugation at 20,000×g, for 2 hours). The former fraction was alble to oxidize succinate, L-lactate, and formate with a comparatively fast rate (Qo_2 (N) of 464, 1150, and, 126 respectively, taking the first 10 minute rates); L-matate and ethanol were oxidized only slowly while L-aspartate, L-glutamate, L-alanine, citrate and fumarate were practically not oxidized. All the mentioned substrates with the exception of citrate and alanine could be oxidized by the non-sedimentable fraction in absence of any acceptor dye. On mixing the two fractions three different kinds of results could be noted depending on the substrates used. (ⅰ) The rate of oxygen consumption of the mixture was approximately equal to the sum of the rates of the individual fractions. This was found for succinate, formate, and L-malate. (ⅱ) Mixing of the two fractions produced a markedly higher rate of the oxidation than what could be accounted for by the activities of individual ones. This is the case for fumarate, L-glutamate, L-aspartate, L-alanine, and ethanol. (ⅲ) The combination of the two fractions lowered the rate of oxygen uptake. This was noted for L-lactate.

本文報告了一種簡便高效的細菌磨,其主要構成部分包括一段横置的磨沙硬質滾筒和凹面瓷座。用5,000×g轉速,20分鐘離心收集的大腸桿菌漿作試驗。這種菌漿含水量約為乾菌重的5倍。加入1.3—1.5倍重的玻璃粉(顆粒小於3μ),混合均匀,研磨20—30秒,細菌裂碎率保證在99.99%以上。 將研磨所得混合物懸浮於磷酸鹽緩衝劑中,離心分離得到全提取液。全提取液再以20,000×g轉速,2小時離心分離,將酶劑分為澄清提取液和從碎菌沉澱製成的碎菌懸浮液兩部分。後者所殘留的活菌的氮約佔全氮的10~(-3)—10~(-4)%。利用同一批培養出來的細菌各次研磨製得的酶劑,其酶活力的可重複性程度很高,以琥珀酸氧化酶系的活力來說,各次平均離差範圍僅±10%。不需要加任何輔酶輔基,這兩部分酶劑都能够直接利用空氣中的氧來氧化琥珀酸、L-蘋果酸、L-乳酸、甲酸。澄清提取液部分還可以氧化L-谷氨酸、L-門冬氨酸、延胡索酸和乙醇,並且當它和碎菌部分酶劑混合時,除琥珀酸、甲酸和L-乳酸外對其他底質的氧化活力都大為提高。
