The present investigation was undertaken in 1965-1966 to study the oxidative phosphorylation and the fine structure of the flight muscle mitochondria in the housefly Musca domestica vicina Macq. Comparisons were made between the normal (= susceptible) and DDT-resistant strains before and after DDT treatment in order to analyse the relationship between structure and function of the mitochondria. The present investigation was undertaken in 1965-1966 to study the oxidative phosphorylation and the fine structure of the flight muscle mitochondria in the housefly Musca domestica vicina Macq. Comparisons were made between the normal (= susceptible) and DDT-resistant strains before and after DDT treatment in order to analyse the relationship between structure and function of the mitochondria. The results may be summarized as follows:1. The mitochondrial suspensions prepared from the flight muscles oxidized rapidly a-glycerophosphate and succinate, but slowly or did not oxidize most of the other citric acid cycle intermediates such as pyruvate, a-ketoglutarate, malate and fumarate as well as β-hydroxybutyrate and glutamate. The same results could be obtained when the muscle homogenate or the mitochondria plus supernatant were tested. This result may serve as the evidence in favour of the view that the isolated housefly mitochondria are not readily permeable to these intermediate compounds of citric acid cycle.2. In the cases where a-glycerophosphate was used as the substrate, a high rate of respiration (Qo2 = 413.3) and a high yield of oxidative phosphorylation (P/0 -2.00) may be obtained in the following incubation conditions: the dilute mitochondrial suspension (about 2 mg of protein of mitochondria in 2.0 ml reaction mixture), the short incubation time (about 10 minutes), and the addition of bovine plasma albumin.3. The rates of respiration and oxidative phosphrylation of the mitochondria were significantly inhibited by DDT in vitro and in vivo, but the degree of inhibition was different in normal and DDT-resistant houseflies. In the mitochondria from the normal strain the inhibition of respiration and that of phosphorylation caused by DDT (0.1 mM) in vitro were 40% and 60.9% respectively, whereas in the DDT-resistant strain the corresponding values were 23.4% and 42.7%. AVhen bovine plasma albumin was added into the incubation medium, the inhibition of respiration and that of phosphorylation of the normal housefly mitochondria were 71.1% and 94.8%, whereas in the DDT-resistant houseflies the values were 59.7% and 87.1% respectively. When the houseflies were treated with DDT (0.5%) suspension in vivo, the inhibitions of respiration and phosphorylation in the normal housefly mitochondria were 21% and 33%, whereas in DDT-resistant houseflies the values were 11.5% and 26.9% respectively. After the addition of bovine plasma albumin into the reaction medium the inhibitions of respiration and phosphorylation in the normal housefly mitochondria were 7.0% and 6.1% respectively, whereas in DDT-resistant houseflies no inhibition was observed. These results indicated that the DDT-resistant houseflies in some degree exhibited at the level of mitochondrial function more resistance to the action of DDT than the normal strain.4. An electron microscopic examination on the fine structure of the flight muscle mitochondria from the two strains of the housefly also confirmed that at the level of the cellular organelles the DDT-resistant houseflies were more resistant to the action of DDT. After the insects were treated with DDT in vivo, the mitochondra from the normal strain showed marked lysis, but in the DDT-resistant strain only some mitochondria were slightly damaged. The result demonstrated clearly that in the housefly the changes in structure and function of the mitochondria were correlative during the development of insecticide resistance.

A comparative study on changes in ATPase activity at plasmallemma and tonoplast of winter wheat seedlings and spring wheat seedlings during cold hardening was carried out by use of cytochemical method of lead phosphate precipitation at ultrastructural level.The following experimental results have been obtained: 1.When the tissue slices from unhardened winter wheat seedlings were incubated for 2.5 hr at 22℃,the heavy lead phosphate deposits indic-ating a high activity of ATPase were obviously localized at plasmallemma,and plasmodesmata,while they were not found at tonoplast and other organ-elles (Fig.1,2).At 3℃ incubation,the ATPase activity at plasmallemma and plasmodesmata of these tissue slices was inhibited.When the tissue slices from cold hardened winter wheat se-edlings were incubated at 22℃ or 3℃,ATPase activity not only was showed at plasmallemma and plasmodesmata,but also at tonoplast (Fig.3,4). 2.When the tissue slices from non-hardened spring wheat seedlings were incubated at 22℃ or 3℃,the ATPase activity reaction of their plas-mallemma and tonoplast was similar to that of unhardened winter wheat tissues under same incubation conditions.Plasmallemma ATPase activity was ob-served at 22℃ incubation,but not or insignificant at 3℃.The ATPase ac-tivity at tonoplast was absent at either 22℃ or 3℃ incubation (Fig.5,6,7).In cold hardened spring wheat tissue slices,the ATPase activity at plasmal-lemma and tonoplast was significantly different from that of cold hardened winter wheat seedlings,Exept plasmal-lemma ATPase activity was present at 22℃ incubation,ATPase activity was not shown at tonoplast at either 22℃ or 3℃ incubation (Fig.9,10). 3.The above results indicated that during cold hardening of winter ??wheat seedlings,the cold-tolerant char-acter of plasmallemma ATPase activity was developed,and so it is allowed to maintain its high activity under low temperature favouring to hardening;and the high ATPase activity at tono-plast was formed.Under hardening of low temperature the cold resistance of spring wheat seedlings is not developed,and the ATPase activity at plasmallemma and tonoplast does not produce the adaptive changes being similar to those of winter wheat mentioned above.Therefore,it is evident that the adaptive changes in ATPase activity at plasmal-lemma and tonoplast of winter wheat during cold hardening are associated with the development of cold resistance of plants.

采用磷酸铅沉淀的电镜细胞化学方法,对比观察了耐寒的冬小麦幼苗和不耐寒的春小麦幼苗在抗寒锻炼过程中质膜及液泡膜ATP酶活性的变化,结果指出:冬小麦质膜ATP酶在抗寒锻炼过程中获得了耐低温的特性,以致在适于锻炼的低温(3℃)下,能保持高的活性反应;同时液泡膜在锻炼过程中形成高的ATP酶活性。春小麦幼苗在低温锻炼中不能提高抗寒力,其质膜ATP酶在锻炼过程中也不能发展抗寒性能,因而在3℃低温培育下不表现明显的活性;同时液泡膜也不产生ATP酶活性。这些结果进一步证明,在抗寒锻炼中,冬小麦质膜耐低温特性的形成,以及在液泡膜上产生高的ATP酶活性,是与植株抗寒力的提高有密切关系的重要特性。

TWO strains of nifogen fixing bacteria, Klebsiella pneumoniae K-12 and Enterobscter sp. E-25, isolated from the rhizosphere of local paddy fields were examined for their associative nitrogen fixation with rice seedlings cultured on C and N free media, in comparison with two known strains Azos-Pirillum lipoferum FS and Klebsiella oxytaca NG-13 introduced from abroad. All of the above strains could grow and possess associative nitrogen fixation activity under dark incubation conditions at 28?. The nitrogen fixing activities of these associations were as follows. 10.5 nmol C2H4/seedling/hr. for Azospirillum Hpoferum FS,4.9 nmol C2H4/seedling/hr.for Klebsiella pneumon-iae K-12, 3.9 nmol C2H4/seedling/hr. for Klebsiella oxytoca NG-13, and 2.9 nmol C2H4/seedling/hr. for Enterobacter sp. E-25 respectively. With the exception of the aerobic Azospirillvm lipofervm FS, the other 3 strains grew vigerously around the endosperm of the seedling and formed a bacteria ring. By using DNA specific flourescenCe dye DAPI staining and scanning electron microscopic observations revealed that these 4 strains of the nitrogen fixing bacteria adhered to the root surface and built up a dense Population, Particularly at the root hair region. Scanning electron microscopic observation of the transverse section of the root and the re-isolation experiments of the endophytes indicated that the bacteria could invade the Parenchyma of the root cortex and propagate within the cell.
