AIM: To raise antibody of rat brain nitric-oxide synthase (bNOS) through immunizing animal with a peptide of bNOS that can represent the holoprotein. METHODS: The amino acid sequence for the bNOS was analyzed by GenePro computer program. According to the hydrophilicity, hydrophobicity, antigenicity, and the potentiality to form protein second structures of α-helix, β-sheet and β-turn, the structure of bNOS was predicted. The peptide 277 - 287 was selected that was predicted to be in the antigen epitope of bNOS....

AIM: To raise antibody of rat brain nitric-oxide synthase (bNOS) through immunizing animal with a peptide of bNOS that can represent the holoprotein. METHODS: The amino acid sequence for the bNOS was analyzed by GenePro computer program. According to the hydrophilicity, hydrophobicity, antigenicity, and the potentiality to form protein second structures of α-helix, β-sheet and β-turn, the structure of bNOS was predicted. The peptide 277 - 287 was selected that was predicted to be in the antigen epitope of bNOS. The peptide was chemically synthesized, coupled to keyhole limphet hemocyanin carrier protein and injected into rabbits to raise antibody. The specificity of the antibody was tested by enzyme-linked immunosorbent assay, immunohisto-chemistry, and Western blotting. RESULTS: The antibody bound the protein in rat cerebellum extract. In Western blotting, the antibody bound the protein band of 150 kDa in SDS-PAGE, and the binding was inhibited by peptide conjugated with carrier protein. In immunohistochemistry, the stain was collocated with the stain in NADPH-dehydrogenase histochemistry. CONCLUSION: The antibody against the peptide recognized the natural bNOS in rat brain, and the peptide 277 -287 was located on the surface of bNOS.

The effects of cerebellum extracts (CE) from different embryonic ages on cultured rat spinal cells were studied. CE from 14dt16d , 18d >20d embryonic rats(E14 - 20) were made into different concentrations on protein,then were added to cultured spinal cells of E18 rats. The neuronal survival was determined by the auto-colorimetric assay. The result showed that CE of all experimental ages could promote the survival of spinal neurons,and there was a concentration-dependent relation on protein, with significant differencesC P <0. 001) between the control and experimental groups. The most prominent neurotrophic activities were found in E14 CE of 0. 20mg/ml and E18 CE of 0. 40mg/ml.

为探讨不同胎龄大鼠小脑提取液(CE)对体外培养脊髓神经细胞的影响,制备不同蛋白浓度的14d、16d、18d和20d胎鼠(E14,E16,E18,E20)CE,加入原代培养的E18胎鼠脊髓神经细胞中,24h后以微量快速自动比色法检测细胞活性。结果显示,实验各胎龄CE均能促进培养脊髓神经元的存活,并有蛋白浓度依赖关系,与对照组及其它各实验组相比有极显著性差异(P<0.01),E14和E18CE分别在蛋白浓度为0.20mg/ml和0.40mg/ml时有最大神经营养活性。结果提示CE对培养脊髓神经细胞具有明显的营养作用,为利用胚胎小脑移植修复脊髓损伤提供了实验依据。
