1. The alkaloid contents of Chinese gelsemium (G. elegans Benth.) have been reinvestigated. Various procedures of isolation of the alkaloids were compared and a more efficient one developed. 1. The alkaloid contents of Chinese gelsemium (G. elegans Benth.) have been reinvestigated. Various procedures of isolation of the alkaloids were compared and a more efficient one developed. The unconventional formula of kouminidine, C_19H_25O_4N_2, is revised to C_20H_26N_2O_4.Preliminary considerations based on UV and IR spectra suggest that kouminidine is possibly an indolenine type alkaloid with a zwitterionic structure.2. The constitution of koumine, the main alkaloid of Kou-Wen, has been preliminarily studied on spectral as well as on chemical ground. It is proposed that, in contrast to gelsemine, koumine probably possesses a pseudoindole ring system. Although gelsemine and koumine both contain an exo-vinyl, an Nb-methyl and most likely a cyclic ether linkage, the structures of the alicyclic parts of these two alkaloids appear also different from each other.

About ten constituents of the bovine thymic hormones have been isolated and purified in various laboratories. Thymosin F_5, the partially purified bovine thymic hormone by A. L. Goldstein et. al. has been successfully used to treat the primary immunodeficiency diseases and clinical application for autoimmune disorders and cancer are also under way in various laboratories. However, there are no detailed studies on the porcine thymic hormones had been reported thus far. In this paper, we report a successful method for the preparation of thvmic hormone from thymus tissue of pig. The procedure is as follows: The crude extract of pig thymus was partially denatured by heat, followed by ammonium sulfate fractionation and finally purified by DEAE-cellulose Chromatography. Our partially purified thymic hormone was free from carbohydrates and nucleotides and contains at least 8—9 protein components, with molecular weight from 9000 to 68000. and Their isoelectric points ranging from pH 5.0 to 7.5. The thymic hormone F_5 thus prepared exhibits very high biological activity as shown by the micro-rosette assay. Data showed that even when this thymic hormone was lower down to O. 1 μg per 1~2×10~5 lymphocytes, it still expresses significantly function to induce the maturation of T-cell populations. The purification method described here is simple and efficient, suitable to prepare the hormone in large scale. Evidence showed that the porcine thymic hormone may be useful in treatmant of primary immunodeficiency, autoimmune disorders, certain cancer diseases and various infections of bacteria viruses and fungi. We have treated some patients with our porcine thymic hormone 4mg/day, 3 times/week im. for periods of one to three months duration. The T-cell percentage of all the patiants apparantly increased, neither as anaphylaxis, nor rash during the treatment period was observed. The activity of our porcine thymic hormone preparation, so far as its biological activity estimated by the rosette bioassay in vitro is concerned, is no lets than that of the bovine thymosin F_3.

本文提出了一个通过部份热变性—硫酸铵沉淀—DEAE纤维索层析等步骤制备猪胸腺激素的方法。所得制品主要是一个由8—9种中性和酸性蛋白质所组成的蛋白混合制剂;分子量从9,000—68,000;不含糖类或核苷酸;具有很高的生物学活性,用微量玫瑰花结法测定,0.1微克蛋白质仍有明显的促使T淋巴细胞成熟的功能。
