Objective Some research has shown that human cytomegalovirus (HCMV) can inhibit the proliferation of marrow progenitors in vitro. This study aims at investigating the effect of HCMV on proliferation of multipotential hematopoietic progenitors (CFU Mix) and exploring the interfering effect of ganciclovir and astragalus. Methods Three infection groups were established by adding HCMV AD 169 solution of different concentrations (1∶10, 1∶100, 1∶1000) into the culture system in vitro. Another group which did not receive any viral solution was used as the control. The effect of HCMV AD 169 strain on cord blood CFU Mix was observed by colony forming unit assay. PCR was used to detect the existence of HCMV DNA in the colony cells of cultured CFU Mix. The suppression effect of HCMV AD 169 on CFU Mix proliferation was interfered by adding ganciclovir and/or astragalus membranaceus to the 1∶10 HCMV infection group. Results (1) The numbers of CFU Mix colonies in the three HCMV AD 169 infection groups were significantly less than those of the Control group. The number of CFU Mix colonies decreased with the increase of HCMV concentration. The suppression effect of HCMV AD 169 on CFU Mix proliferation also showed a dose dependent way: 55.2% inhibition rate in the 1∶10 HCMV infection group, 35.1% in the 1∶100 HCMV infection group and 20.3% in the 1∶1000 HCMV infection group. The peak time of CFU Mix colonies in the three viral infection groups did not significantly differ from the Control group, but the lasting time of the CFU Mix colonies in the infection groups was significantly shorter than that in the Control group [(15-18) d vs (22-24) d); (P< 0.01 ). (3) HCMV DNA was positively detected in the colony cells of the three viral infection groups, while negatively in the Control group. (4) The number of CFU Mix colonies in the 1∶10 HCMV infection group increased significantly after gancicovir and or astragalus membranaceus administration. [WTHZ]Conclusions The HCMV AD 169 strain can inhibit the proliferation of CFU Mix. Ganciclovir and astragalus membranaceus may have an anti HCMV effect.

Objective: To investigate the effect of human cytomegalovirus (HCMV) on proliferation of megakaryocyte progenitors (CFU-Mk) and explore the interfering effect of astragalus membranaceus(AM) on the effect induced by HCMV in vitro. Methods: The effect of HCMV-AD169 strain on cord blood CFU-Mk was observed by Colony forming unit-assay. PCR and fluorescence quantification PCR were used to detect the existence of HCMV-DNA in the colony cells of cultured CFU-Mk. The suppression effect of HCMV on CFU-Mk was interfered in by adding AM in 10 -1 HCMV group. Results: (1) The numbers of CFU-Mk colonies decreased significantly with HCMV-AD 169 infection compared with the controls, and showed an increase tendency with the decrease of HCMV consentration. (2) The peak time of CFU-Mk colonies in the viral groups did not significantly differ from the control groups, but the lasting time of the CFU-Mk colonies in the viral groups was significantly shorter than that in control groups [(22~27)d vs (28~30)d,P<0.01]. (3) HCMV-DNA was positively detected in the colony cells of viral groups by PCR, while negative in the control groups. HCMV-AD169 DNA copies decreased significantly by fluorescence quantification PCR in 10-1HCMV group with the presence of AM. (4)The numbers of CFU-Mk in 10-1 HCMV group increased significantly with the presence of AM. Conclusion: HCMV-AD169 strain can inhibit the proliferation of CFU-Mk by infecting the hematopoietic progenitors, which may be the main reason of HCMV infection associating with thrombocytopenia. AM may have direct effect of anti-HCMV in vitro.

目的 探讨人巨细胞病毒 (HCMV)对脐血巨核系祖细胞 (CFU -Mk)体外增殖的影响及黄芪对此的干预作用。方法 采用造血祖细胞体外半固体培养技术 ,观察、计数HCMV感染的CFU -Mk集落数、抑制率、集落峰值及维持时间 ;并用定性和定量PCR检测集落细胞内HCMV -DNA ;在 10 - 1感染组中加入黄芪后 ,观察黄芪对HCMV抑制CFU -Mk集落生长的干预作用。结果 ①HCMV感染组CFU -Mk集落数较对照组明显减少 (P <0 .0 1) ;②各组集落峰值出现时间无明显差异 (P >0 .0 5 ) ,但感染组集落维持时间较对照组明显缩短 (P <0 .0 1) ;③用PCR技术在感染组集落细胞内检测到HCMV -DNA的存在 ;④黄芪组CFU -Mk集落数明显提高 ,荧光定量PCR显示集落细胞内HCMV -DNAcopies较 10 - 1感染组明显减少。结论 在体外HCMV -AD169对CFU -Mk的增殖和分化有明显抑制作用。HCMV能直接感染巨核系祖细胞 ,此作用可能与临床HCMV感染出现血小板减少有关 ;黄芪在体外有明显的抗HCMV活性。
