Results The motor nerve conductive speed in group E was 38.23(m/s) and there was no significant difference compared with group D ( P >0.05),but better than group B and C.The mixed lymphocyte culture in group E was 2 668.37(cpm).The delayed type hypersensitivity in group E was 0.510(cpm).The detections in histology and electron microscope confirmed that group A was better than group E and group E was better than group B,C and D. Conclusion The intrathymical injection of donor MHC antigen could induce splecific immunotolerance of sciatic nerve transplantation.

目的 探讨胸腺内注射异基因抗原对异体鼠坐骨神经移植存活的影响。 方法  70只Balb/c小鼠随机分为 5组 :自体神经移植组 (A组 )、异体神经移植组 (B组 )、冷冻后异体神经移植组 (C组 )、应用免疫抑制剂异体神经移植组 (D组 )和胸腺内注射供体组织相容性 (MHC)抗原后移植组 (E组 )。 3周后进行神经电生理检查、组织学检查、混合淋巴细胞培养及迟发性超敏反应的检测。 结果 运动神经传导速度E组为 3 8 2 3 (m /s) ,与D组相比差异无显著性 (P >0 0 5 ) ,且E组优于C组、B组。混合淋巴细胞培养E组为 2 668 3 7(cpm ) ,迟发性超敏反应E组为 0 5 10 (cpm ) ,组织学及电镜检查均证实A组优于E组、E组分别优于C组、D组、B组。 结论 胸腺内注射异基因抗原可诱导对异体坐骨神经移植的特异性免疫耐受。

Objective:To prepare a new nerve extracellular matrix(ECM)for the recovery of segmental defects of the peripheral nerve in rabbits. Methods:The materials were made by immersing the samples of sciatic nerve into NaOH aqueous solution and cleaned by ultrasonic wave(USW),then observed it by scanning electron microscope(SEM).The materials were implanted into the muscles of rabbits for the study of biocompatibility, and then bridged to repair segmental defects of the peripheral nerve in rabbits....

Objective:To prepare a new nerve extracellular matrix(ECM)for the recovery of segmental defects of the peripheral nerve in rabbits. Methods:The materials were made by immersing the samples of sciatic nerve into NaOH aqueous solution and cleaned by ultrasonic wave(USW),then observed it by scanning electron microscope(SEM).The materials were implanted into the muscles of rabbits for the study of biocompatibility, and then bridged to repair segmental defects of the peripheral nerve in rabbits. Under light microscope, diameter of myelinated nerve fibers and axis-cylinder in the grafts were examined with HE staining at different postoperative intervals (8 and 16 weeks). In addition, Nerve electrophysiology was recorded at 16 weeks after transplantation. Results:(1) The cellular elements of the sciatic nerve treated with NaOH-maceration were eliminated completely, while the original three-dimensional structure of the endoneurium, perineurium and epineurium were preserved integrately. (2) No significant difference was observedbetween the experiment groups and the control groups ( P >0.05) were checked up. Conclusion:The ECM has a good biocompatibility, which can induce nerve fibers to regenerate. This study provide an experiment foundation for clinical nerve surgery.

目的 :制备一种新型天然神经细胞外基质材料 ,通过动物实验研究 ,探讨其修复周围神经干节段性缺损的可行性。方法 :采用NaOH消蚀法 ,制备家兔坐骨神经细胞外基质材料 ,行扫描电镜观察及生物相容性实验 ,用于桥接修复家兔坐骨神经节段性缺损并以自体神经修复作为对照 ,术后通过肌电图检测、再生神经纤维组织学观察等方法 ,证实该支架可有效地引导和促进神经纤维再生。结果 :(1)经NaOH消蚀处理的坐骨神经组织 ,其细胞成分被完全消蚀掉 ,神经膜管保持原有的构筑特征 ;无明显排异反应并可降解吸收。 (2 )该材料和自体神经移植修复周围神经干节段性缺损在电生理和组织结构的恢复方面经统计学处理无显著性差异 (P >0 .0 5 )。结论 :此方法制备的细胞外基质材料可有效地桥接修复周围神经干节段性缺损 ,为其临床应用奠定实验基础。

Objective To evaluate the effects of bridging by skeletal muscle(semi-anastomsois and semi-implantation method)and nerve graft on defect of the peripheral nerves,discuss the use of semi-anastomsois and semi-implantation method in clinic. Objective To evaluate the effects of bridging by skeletal muscle(semi-anastomsois and semi-implantation method)and nerve graft on defect of the peripheral nerves,discuss the use of semi-anastomsois and semi-implantation method in clinic. At the same time, right common peroneael nerves were treated by the nerve graft, the changes of the two sides were measured after three months. Results After three months,the functions of two legs resumed.By macrography and observing under microscope,nerve electrophysiology,histology, there was no significant difference between two groups in the number regenerated fiber crossed sectional area. The effects of semi-anastomsois and semi-implantationwith skeletal muscle was as same as nerve graft. Conclusion The method of semi-anastomsois and semi-implantation with skeletal muscle not only operates easily but also saves time and inosculates reliably.It would be a potential method on restoring nerve defect .It has applicable value.

目的:比较用半吻合、半埋入法骨骼肌桥接与神经移植治疗神经缺损的效果,以探讨半吻合、半埋入法骨骼肌桥接的临床应用价值。方法:选用体重200~300g成年Wistar大白鼠15只,随机选择每只左侧后肢的坐骨神经造成的神经缺损用半吻合、半埋入法骨骼肌桥接,右侧神经缺损采用神经束膜缝合移植修复。大白鼠分笼饲养3个月。结果:3个月后,大白鼠双下肢功能均得到恢复,通过肉眼观察、手术显微镜下观察、神经肌电图检查及组织学检查,骨骼肌桥接侧与神经移植侧差异无显著性。半吻合、半埋入法骨骼肌桥接治疗周围神经缺损取得与神经移植相近的结果。结论:半吻合、半埋入法骨骼肌桥接手术难度低、时间短、吻合牢靠,是一种具有潜在临床应用价值的神经修复方法。
