In order to find a,simplified but dependable method for active immunization against swine erysipelas and hog-cholera,a mixed bivalent vaccine was triec and the results may be briefly summarized as follows: (1) This vaccine was composed of an attenuated strain of Erysipelothrix rhusiopathiae mixed with a lapinized hog-cholera virus in a definite proportion,suspended in diluted milk powder solution and 1% sucrose,and finally lyophilized. (2) The mixed bivalent vaccine was diluted with 1.2% Al(OH)_3 gel,andi 2 ml of this vaccine were administered intramuscularly for pig immunization resulting with 100% immunity to both diseases at the end of 7 months and 75% to hog-cholera and 100% to swine erysipelas at the end of 9 months. (3) The keeping quality of the vaccine was tested and it was found that excellent antigenicity was still maintained at the end of 6 months under a temperature condition varying from-8℃ to,35℃. (4) Safety test of the vaccine was carried out both in laboratory and out in the field,and a total of 16,374 pigs of different breeds at various ages was used for the test with only 0.22% of the total animals showing some vaccination reactions but no mortality.A general survey was made 7 months after vaccination and neither swine erysipelas nor hog-cholera cases were found among tested animals.

Two strains of Capromyces Pleuropneumoniae were propagated in and passed through embryonated eggs for about 80 generations by means of yolk-sac inoculation. Both the strains retained their full virulence in the first 5-6 generations as titrated in goats. Formalized vaccine prepared from the embryonated tissue of the first 3-7 generations combined with aluminum hydroxide gel did not protect all of the goats vaccinated from artificial challenge. However, by continuously passing through embryonated eggs, both of the strains gradually lost their infectivity for goats. The virulence of one of the strains on the 17th passage, and the other on 55th passage decreated to such an extent that even intratracheal injection with 1 gram of yolk or whole egg virus failed to produce any clinical symtoms in goats, whereas the minimum infectious dose of the stock lung tissue virus for goats was 0.000004 gram only. Thirty-two goats were inoculated subcutaneously with 0.25-1.0 gram of these virus containing materials, but none of them showed thermal or local reactions; on the other hand, 31 of them engendered solid immunity to artificial challenge. It must be noted that the attenuated virus seems to liable to lyophilization, the dosage of the lyophilized tissue virus required to confer immunity in goats is much larger than that of wet tissue virus. In a field test, several flocks of pregnant ewes were vaccinated with whole egg tissue virus of one the strains on its 63rd generation. It was surprising to see that although no thermal or local reaction was observed, most of the ewes aborted following injection.

Summary In order to find a simple and rapid method for evaluation of swine erysipelas serum, the hemagglutination-inhibition test was tried and the successful results may be briefly summarized as follows: (1) Among more than one hundred strains of Erysipelothrix rhusiopathiae under test, a particular strain, E. 5601, was found to agglutinate chicken erythrocytes more clearly than others in the same way as described for influenza virus. (2) The agglutinating action of strain E. 5601 was specifically inhibited by swine erysipelas immune sera, and was not influenced by normal serum, hog-cholera serum, nor by hemorrhagic septicemia (or pasteurellosis) sera. (3) Thirteen batches of swine erysipelas hyper-immune sera were detected for their potency, and, as a result, it was found that the hemagglutination-inhibition titers of different batches showed a series of gradient degrees in which potent sera always possessed an inhibition titer above 1: 384, impotent ones usually below 1:192 or less, and normal serum or sera other than swine erysipelas almost no titer. This fact was confirmed by a series of correlated biological tests with mice. (4) By using this method and trying to get ready all those necessary materials and equipments on hand, the potency of swine erysipelas hyper-immune sera can be evaluated within on hour.

一、通过猪丹毒杆菌有与鸡血球凝集的特性,笔者自100多个猪丹毒杆菌品系內找出了凝集作用良好的猪丹毒杆菌E、5601品系,并用来供制本試驗的抗原。二、本試驗用的抗猪丹毒免疫血清共有13个批号,并以健馬血清、抗出敗血清等四种血清作了对照試驗。試驗結果,各个抗丹毒免疫血清的凝集抑制价高低表現不一,有凝集抑制价甚低者,有高达1:384者。一般在有效期內的抗猪丹毒免疫血清,其凝集抑制价最高;失效或距失效期較久的血清則凝集抑制价逐漸减低。至健馬血清、抗禽畜出敗血清等非特异性血清均无凝集抑制現象。三、小白鼠作血清效力对照試驗說明:鸡血球凝集抑制价的高低与抗猪丹毒血清的免疫效力有直接的密切关系,經以13批抗猪丹毒血清几次重复試驗証明,无凝集抑制价的各类血清或者凝集抑制价低于1:192者的抗猪丹毒血清,对小白鼠的保护力亦差;若凝集抑制价能达1:384者,則血清效力佳良,并可符合国家頒布的效力标准要求合格适用。四、本报告內就猪丹毒免疫血清鸡血球凝集抑制試驗的操作方法以及在猪丹毒免疫血清生产上和血清效力鑑定上的实用价值,均經加以叙述与闡明。
