Objective To observe the bio-effects on VEC-304 treated with low-frequency ultrasound and micro bubble agent,and to offer the clinic application theory basis of ultrasound.Methods Cells were divided into four groups :control(A);pure microbubble agent(B);pure ultrasound(C),according to the exposure time,group C was subdivided into C_(1)(exposure 60()s),_()C_(2)(exposure 90()s),C_(3)(exposure 120()s) and C_(4)(exposure 150()s);low-frequency ultrasound combined with micro bubble agent(D),according to the exposure time,group D was subdivided into D_(1)(exposure 60()s),D_(2)(exposure 90 s),D_(3)(exposure 120 s) and D_(4)(exposure 150()s).The survival rate of VEC-304 was detected by MTT colormetric assay.Visible spectrophotometric was applied to determine ROS and SOD.Apoptosis was analyzed by flow cytometry after ultrasonic exposure.Results The proliferation of group D were obviously inhibited,which was detected by MTT colormetric assay.The yield of ROS of groups C_(3) and D_(3) was higher than groups A and B,respectively.The yield of SOD of groups C_(3) and D_(3) was lower than groups A and B,respectively.The apoptosis rates were found to reach(11.57±2.68)% in group C_(3) and(21.15±4.68)% in group D_(3) by FCM.Conclusion Low-frequency ultrasound combinding with microbubble agent can induce VEC-304 injury and apoptosis,which is probably due to the adding yield of ROS in cultured liquid.

目的:研究低频超声联合微泡剂对血管内皮细胞VEC-304的作用,为超声治疗的临床应用提供实验依据。方法:将细胞分为空白对照(A)组;单纯微泡剂(B)组;单纯超声(C)组,因给予不同时间的超声辐照又分为C1组(60 s)、C2组(90 s)、C3组(120 s)和C4组(150 s);超声联合微泡剂(D)组,因加入微泡剂后立即给予不同时间的超声辐照又分为D1组(60 s)、D2组(90 s)、D3组(120 s)和D4组(150 s)。MTT法观察细胞增殖的抑制作用,分光光度法检测细胞培养液中活性氧(ROS)和超氧化物歧化酶(SOD)活力的变化,流式细胞仪(FCM)检测细胞凋亡。结果:超声联合微泡剂能够显著抑制细胞增殖;C3、D3组培养液中ROS活力显著高于A、B两组(P<0.01),SOD活力显著低于A、B两组(P<0.01),且D3组效应比C3组更为明显;C3、D3组细胞凋亡率明显高于A、B组(P<0.01),D3组高于C3组(P<0.05)。结论:低频超声联合微泡剂可能是通过增加细胞外ROS的活力来诱导对细胞的损伤和凋亡。

Objective: To explore the effect of low power ultrasound on ovarian angiogenesis of rabbits.Methods:Forty rabbits received CO2 micro-bubble agent injection through one ear peripheral vein,after which their abdomen were radiated with low power ultrasound(0.5W),according to radiation time,animals were divided into 4 experimental groups and 1 control group.A group received radiation for 30 seconds,B group was 60 seconds,C group was 120 seconds and D group was 4 minutes,control group was not radiated.After the radiation,4 rabbits of every group were anatomized,their double ovaries were fixed with formalin,ovarian angiogenesis embolization was observed with HE.Other rabbits were breeded for one month before ovarian angiogenesis were observed in the same methods. Results: After the radiation,four rabbits of every group were anatomized in 24 hours,thrombus could be seen in the ovaries of the four groups,but they were few.The thrombus formation ratio of every group were(3.67±0.83)%?(3.54±1.07)%?(3.39±1.09)% and(3.92±1.10)% respectively.Compared with the controlled group,the result from the experimental group was not significant(P>0.05);After one month,thrombus could not be seen in the ovaries of all four groups.No injury could be seen in all kinds of cells in the four groups. Conclusion:Rabbit ovarian angiogenesis was not affected by low power ultrasound.

目的:探讨低功率超声辐射微泡剂对家兔卵巢微血管的影响,为该方法在临床上的进一步应用提供理论依据。方法:40只生育期的家兔随机分为四个实验组和一个对照组,每组8只家兔。由家兔一侧耳缘静脉注射微泡剂,然后以低功率低频率(0.5W)超声照射家兔腹部的4个象限,按照射时间长短分为30s组,60s组,120s组和240s组。照射结束24h后解剖四个实验组和对照组各4只家兔,取双侧卵巢,10%甲醛固定后进行苏木精-伊红(H-E)染色,观察家兔卵巢微血管内血栓形成情况。各组另4只家兔继续饲养,30d后解剖取双侧卵巢。结果:超声干预后24 h解剖的四个实验组家兔,双侧卵巢的微血管内偶可见血栓,其血栓形成率分别为(3.67±0.83)%、(3.54±1.07)%、(3.39±1.09)%和(3.92±1.10)%,各组间比较无显著性统计学差异(P>0.05)。而超声干预后饲养30d解剖的4个实验组家兔卵巢微血管内均未见明显的损伤。各组卵巢内的细胞饱满,无细胞核碎裂、固缩等细胞死亡征象。结论:本实验结果可初步认为低功率超声辐射微泡剂对家兔卵巢正常微血管似无明显影响。
