Objective To investigate the effects of chitooligosaccharide on the lactate dehydrogenase and γ-glutamyltransminase of Streptococcus mutans . Methods Streptococcus mutans ATCC 25175 was chosen as the experimental bacteria. The effects of chitooligosaccharide on the endoenzymes were investigated by observation of the enzyme actitities of LDH and γ-GT in bacterial solution.The data of were analyzed by an analysis of variance (ANOVA).[WTHZ]Results The releasing of LDH and γ-GT was proved.It was found that the enzyme actitities of LDH and γ-GT in bacterial solution increased with the concentration of chitooligosaccharide. Conclusion The chitooligasccharide had a destructive effect on the cell walls of S.mutans.

目的研究壳寡聚糖对变形链球菌乳酸脱氢酶(LDH)及γ谷丙转氨酶(γ-GT)的影响。方法将变形链球菌接种在含壳寡聚糖的培养基中,测定培养上清液中乳酸脱氢酶及γ谷丙转氨酶等细胞内酶的含量。结果菌液上清中的LDH和γ-GT2种细胞内酶含量显著高于空白对照组。表明接种在含壳寡聚糖培养基中的细菌出现了明显的细胞内酶溢出,并与浓度相关。结论壳寡聚糖对细菌的细胞壁有破坏作用,可能是壳寡聚糖具有防龋功能的原因。

Objective:To study the antimicrobial effect of polymethylmethacrylate (PMMA) denture base with silver-containing antimicrobial agents of nanometer level against Streptococcus mutans and Candida albicans in vitro. Methods:Minimum inhibitory concentration (MIC) of the silver-containing antimicrobial agent against S. mutans and C. albicans was examined. According to the MIC, 4 concentrations(1,2,5 and 10 mg/ml) of the agent were selected for the preparation of antimicrobial PMMA resin base. Then, the antimicrobial effect of the resin base was examined by in vitro bacteria culture and the most probable number(MPN) counting. Results:The MICs of the agent against S. mutans and C. albicans were 0.1 mg/ml and 1 mg/ml respectively. With the agent at 1, 2, 5 and 10 mg/ml, the inhibition ratios of the base against S.mutans were 67.4%,71.3%,99.0 and 99.5% respectively, that against C. albicans were 25.8%,54.8%,90.3% and 93.0%, respectively. Conclusion:The polymethylmethacrylate resin base with silver-containing antimicrobial agent of nanometer level at 5 mg/ml has ideal antimicrobial activity against S. mutans and C. albicans.

目的研究纳米载银树脂基托在体外对变形链球菌和白色念珠菌的抗菌效果。方法先测定纳米载银抗菌剂对变形链球菌和白色念珠菌的最小抑菌浓度,并以此为依据在聚甲基丙烯酸甲酯基托树脂粉中添加不同比例的抗菌剂,制成抗菌树脂基托,用贴膜法测定该树脂基托对变形链球菌和白色念珠菌的抗菌率。结果纳米载银抗菌剂对变形链球菌的最小抑菌浓度为0.1mg/ml,对白色念珠菌的最小抑菌浓度为1mg/ml。当添加抗菌剂的浓度分别为1、2、5、10mg/ml时,树脂基托对变形链球菌的抗菌率分别为67.4%、71.3%、99.0%、99.5%;对白色念珠菌的抗菌率分别为25.8%、54.8%、90.3%、93.0%。结论载银抗菌树脂基托在体外表现了一定的抗变形链球菌和抗白色念珠菌的效果,当树脂基托中抗菌剂的浓度达到5mg/ml时,抗菌效果满意。

目的评估三种垫底材料的抑菌活性。方法采用细菌培养琼脂扩散法,将三种垫底材料(3M VitrebondTM玻璃离子垫底材料,CAV-HYCALTM氢氧化钙垫底材料,氧化锌丁香油粘固剂ZOE)置于变形链球菌培养基中,观察各种垫底材料产生的抑菌圈直径的大小,并以此为指标比较三种垫底材料对变形链球菌的抑菌性。结果三种垫底材料对变形链球菌的抑菌作用具有显著性差异,VitrebondTM对变形链球菌具有最强的抑菌性能,其次是CAV-HYCALTM氢氧化钙,ZOE抑菌性能最弱。结论VitrebondTM玻璃离子垫底材料对变形链球菌具有较强的抑制能力,是一种比较理想的垫底材料。
